Encapsulating peptide-stabilized gold nanoclusters (AuNCs) with thiolated hyaluronic acid (HA-SH) and selectively adding cysteine to the peptide sequence increased their photoluminescence. We found that peptide compositions with cysteine in the middle emitted the most. RCR-stabilized AuNCs can be purified using size-exclusion chromatography to characterize their optical characteristics, chemical composition, and possible structure.

The molecular basis of receptor bias in G protein-coupled receptors (GPCRs) caused by mutations that preferentially activate specific intracellular transducers over others remains poorly understood. Two experimentally identified biased variants of β-adrenergic receptors (βAR), a prototypical GPCR, are a triple mutant (T68F, Y132A, and Y219A) and a single mutant (Y219A); the former bias the receptor toward the β-arrestin pathway by disfavoring G protein engagement, while the latter induces G protein signaling explicitly due to selection against GPCR kinases (GRKs) that phosphorylate the receptor as a prerequisite of β-arrestin binding. Though rigorous characterizations have revealed functional implications of these mutations, the atomistic origin of the observed transducer selectivity is not clear.

Gold nanoclusters (AuNCs) are promising nanomaterials for ratiometric fluorescent probes due to their tunable fluorescence wavelengths dependent on size and structure, as well as their biocompatibility and resistance to photobleaching. This study synthesized two azide-functionalized monomers through -dichloro xylene and double-decker silsesquioxane (DDSQ) units with NaN to form DB-N and DDSQ-N monomers, respectively. In addition, five different propargyl-functionalized monomers were also prepared from hydroquinone, bisphenol A, bis(4-hydroxyphenyl)methanone, 2,4-dihydroxybenzaldehyde (then reacted with hydrazine hydrate solution) and 1,2-bis(4-hydroxyphenyl)-1,2-diphenylethene with propargyl bromide to form P-B, P-BPA, P-CO, P-NP, and P-TPE monomers, respectively. As a result, various DDSQ-based main chain copolymers could be synthesized using Cu(I)-catalyzed click polymerization through DDSQ-N with different propargyl-functionalized monomers, of which the chemical structure and molecular weight could be confirmed by using Fourier-transform infrared spectroscopy (FTIR), nuclear magnetic resonance (NMR), and gel permeation chromatography (GPC) analyses.

This study fabricated yellow-emitting CDs (Y-CDs) by hydrothermal treatment of citric acid and urea and applied them as a fluorescence turn-on platform for sensitive and selective detection of lipopolysaccharide (LPS) based on the non-shifted AIEE of peptide-stabilized CD aggregates. The designed peptide (named K3) consisting of aggregation-active and LPS-recognition units triggered the aggregation of Y-CDs, switching on their fluorescence through the blue-shifted AIEE process. The formed K3-stabilized Y-CD aggregates (K3-YCDAs) specifically interacted with LPS at neutral pH, demonstrating that the sequence of the decorated peptide was highly connected with their selectivity and sensitivity.
