Objectives: Multilevel network meta-regression (ML-NMR) leverages individual patient data (IPD) and aggregate data from a network of randomized controlled trials (RCTs) to assess the comparative efficacy of multiple treatments, while adjusting for between-study differences. We provide an overview of ML-NMR for time-to-event outcomes and apply it to an illustrative case study, including example R code.

Methods: The case study evaluated the comparative efficacy of idecabtagene vicleucel (ide-cel), selinexor+dexamethasone (Sd), belantamab mafodotin (BM), and conventional care (CC) for patients with triple-class exposed relapsed/refractory multiple myeloma in terms of overall survival.

A systematic review and network meta-analysis (NMA) was performed to evaluate the efficacy of first-line treatments for locally recurrent unresectable or metastatic triple-negative breast cancer (TNBC) patients. Databases were searched for randomized controlled trials evaluating first-line treatments for locally recurrent unresectable or metastatic TNBC patients. NMA was performed to estimate relative treatment effects on overall and progression-free survival between pembrolizumab + chemotherapy and other interventions.

Article Synopsis
  • * An indirect comparison study was conducted to evaluate the effectiveness of brexu-cel against pirtobrutinib, using patient data to account for various prognostic factors, including disease stage and prior treatments.
  • * Results indicated that brexu-cel led to significantly higher response rates and improved progression-free survival compared to pirtobrutinib, suggesting it might be a more effective treatment option for this patient population.

Cisplatin (-diamminedichloroplatinum(II)) is a potent chemotherapeutic agent commonly used to treat cancer. However, its use also leads to serious side effects, such as nephrotoxicity, ototoxicity, and cardiotoxicity, which limit the dose that can be safely administered to patients. To minimize these problems, dendrimers may be used as carriers for cisplatin through the coordination of their terminal functional groups to platinum.

The development of algorithms for remote sensing of water quality (RSWQ) requires a large amount of in situ data to account for the bio-geo-optical diversity of inland and coastal waters. The GLObal Reflectance community dataset for Imaging and optical sensing of Aquatic environments (GLORIA) includes 7,572 curated hyperspectral remote sensing reflectance measurements at 1 nm intervals within the 350 to 900 nm wavelength range. In addition, at least one co-located water quality measurement of chlorophyll a, total suspended solids, absorption by dissolved substances, and Secchi depth, is provided.

Dendrimers have been studied as promising materials for the delivery of anticancer drugs. In this work, low-generation (0-2) nitrile poly(alkylidenamine)-based dendrimers were explored as nanocarriers for the organometallic complex [Ru(η-CH)(PPh)] (RuCp) and investigated for their anticancer action and involved mechanisms, which were evaluated both and . It was observed that their biological behaviour is generation dependent, where the highest generation metallodendrimer (G2Ru) was overall more effective than the other metallodendrimers.

Background: Overall survival (OS) is the most patient-relevant outcome in oncology; however, in early cancers, large sample sizes and extended follow-up durations are needed to detect statistically significant differences in OS between interventions. Use of early time-to-event outcomes as surrogates for OS can help facilitate faster approval of cancer therapies. In locally advanced head and neck squamous cell carcinoma (LA-HNSCC), event-free survival (EFS) was previously evaluated as a surrogate outcome (Michiels 2009) and demonstrated a strong correlation with OS.

The complexity of drug delivery mechanisms calls for the development of new transport system designs. Here, we report a robust synthetic procedure toward stable glycodendrimer (glyco-DDM) series bearing glucose, galactose, and oligo(ethylene glycol)-modified galactose peripheral units. In vitro cytotoxicity assays showed exceptional biocompatibility of the glyco-DDMs.

Background: Walking speed discriminates levels of functionality in stroke survivors, but its relationship with neuromuscular parameters remains unclear. We aimed to (1) verify relationships between walking speed, peak torque, and rate of torque development in individuals with chronic stroke and (2) investigate whether adjusting the predictive model for assistive device use and lower limb motor function improves its accuracy.

Methods: Twenty-nine stroke survivors (time post-stroke ≥6 months) were assessed for walking speed, motor function, torque of knee extensors, and rate of torque development.

The transport of patients by ambulance is part of the services offered by a Hospital and its importance lies in facilitating the connection between the institutes involved in patient care. However, a significant amount of complaints about irritability and aggressiveness of drivers was registered after a strategic change in the organizational structure that directly affected these professionals. This study aimed to survey the health issues of ambulance drivers at this hospital, from the reorganization of work, through an ergonomic analysis of work.

: This study aimed to assess skin temperature (Tsk) changes after different magnitudes of exercise-induced muscle damage (EIMD) of the elbow flexor muscles and investigate whether Tsk is related to indirect markers of EIMD. : Thirty healthy active men were randomly allocated to the mild (MiDG) ( = 15) or moderate (MoDG) ( = 15) muscle damage group. Muscle damage in elbow flexor muscles was induced by 10 (for MiDG) or 30 (for MoDG) maximal eccentric isokinetic contractions at 60 degrees/s.

: For decades, endocrine therapy has been the cornerstone of management for luminal breast cancer. Despite the substantial benefit derived by patients from endocrine therapy, primary and secondary resistances to endocrine therapy are serious clinical issues.: Today, in the advanced setting, three distinct classes of targeted agents mTOR, CDK 4/6, and PI3K inhibitors, are approved for use.

In this article, a finite element (FE) thermal-electrical model with a trunk-conical discharge channel is employed to simulate individual EDM discharges with a time-on of 18 μs up to 320 μs, which are subsequently compared with the experimental results to validate the model. The discharge channel is a trunk-conical electrical conductor which dissipates heat by the Joule heating effect, being the correspondent factor equal to 1. Instead of the usual copper-iron electrode combination, steel (DIN CK45) and aluminium alloys (DIN 3.

Determination of the chemical composition in the body and carcass of ruminants is important for both nutritional requirement studies and the meat industry. This study aimed to develop equations to predict the body and carcass chemical composition of hair sheep using the chemical composition of body parts, carcass measurements and shrunk BW as predictors. A database containing 107 individual records for castrated male hair sheep ranging from 24 to 43 kg BW was gathered from two body composition studies.

We studied the possibility of detection of [Ru(η-CH)(PPh)Cl] (abbreviated by RuCp) complex as a model system for Ru-based metallodrugs in human urine by using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) without previous purification or removal of inorganic salts. Inorganic salts might prevent the detection of RuCp by MALDI-TOF MS, most likely through the increased number and intensity of background/organic matrix signals. This problem might be overcome by the acquisition of matrix-free spectra and the addition of nanoparticles, such as carbon dots, to the urine solution.

Traditionally, water conditions of coffee areas are monitored by measuring the leaf water potential (ΨW) throughout a pressure pump. However, there is a demand for the development of technologies that can estimate large areas or regions. In this context, the objective of this study was to estimate the ΨW by surface reflectance values and vegetation indices obtained from the Landsat-8/OLI sensor in Minas Gerais-Brazil Several algorithms using OLI bands and vegetation indexes were evaluated and from the correlation analysis, a quadratic algorithm that uses the Normalized Difference Vegetation Index (NDVI) performed better, with a correlation coefficient (R2) of 0.
