View Article and Find Full Text PDFObjective: To provide a feasible and cost-effective next-generation sequencing (NGS) method for accurate identification of viral pathogens in clinical specimens, because enormous limitations impede the clinical use of common NGS, such as high cost, complicated procedures, tremendous data analysis, and high background noise in clinical samples.
Methods: Viruses from cell culture materials or clinical specimens were identified following an improved NGS procedure: reduction of background noise by sample preprocessing, viral enrichment by barcoded oligonucleotide (random hexamer or non-ribosomal hexanucleotide) primer-based amplification, fragmentation-free library construction and sequencing of one-tube mixtures, as well as rapid data analysis using an in-house pipeline.
Results: NGS data demonstrated that both barcoded primer sets were useful to simultaneously capture multiple viral pathogens in cell culture materials or clinical specimens and verified that hexanucleotide primers captured as many viral sequences as hexamers did.

This study aimed to analyze the enzyme activities and gene expression differences related to seed germination in different wheat genotypes, and to clarify the relationship between seed vigor and the related enzyme activities as well as gene expression under stress germination conditions. We measured seed vigor, total soluble sugar content, soluble protein content, α-amylase activity, cysteine protease activity and gene expression of the related enzymes of four wheat cultivars under drought, artificial aging and cold soaking stress. Results showed that drought, artificial aging and cold soaking stress affected seed vigor to some extent.

It was found that Ni-BDC (BDC = 1,4-dicarboxybenzene) can emulsify water and ILs and favor the formation of high internal phase emulsions (HIPEs) under certain experimental conditions. The microstructures of the HIPEs were characterized by confocal laser scanning microscopy using a fluorescent dye Rhodamine B, which proves that the HIPEs are the IL-in-water type.

The results showed that microorganisms from nine genera were related to phosphorus cycle with Bacillus and Pseudomonas being the dominant. The abundance of PSMs was highest in ALS, followed by RPS, RSP2 and BSP2, RSP1 and BSP1, respectively.
