The development of rBAY, a recombinant peptide with the similar sequence of synthetic BAY55-9837, as a potential peptide therapeutic for type 2 diabetes is still a challenge mainly because of its poor stability in aqueous solution. To improve the peptide stability and bioactivity and investigate its biological effects for VPAC2-specific activation, RBAYL with 31 aa was designed based on sequence alignments of pituitary adenylate cyclase-activating peptides (PACAPs), vasoactive intestinal peptide (VIP), and related analogs and generated through sitedirected mutagenesis. Stability analysis showed that the prepared RBAYL with three mutations (N9Q, V17L, and N28K) were much more stable than rBAY.

View Article and Find Full Text PDFA versatile two-step wet process to fabricate Pt, Pd, Rh, and Ru nanoparticle films (simplified as nanofilms hereafter) for in situ attenuated total reflection Fourier transform infrared (ATR-FTIR) study of electrochemical interfaces is presented, which incorporates an initial chemical deposition of a gold nanofilm on the basal plane of a silicon prism with the subsequent electrodepostion of desired platinum group metal overlayers. Galvanostatic electrodeposition of Pt, Rh, and Pd from phosphate or perchloric acid electrolytes, or potentiostatic electrodeposition of Ru from a sulfuric acid electrolyte, yields sufficiently "pinhole-free" overlayers as evidenced by electrochemical and spectroscopic characterizations. The Pt group metal nanofilms thus obtained exhibit strongly enhanced IR absorption.

Methods: The serum concentration of IL-15 was detected using ELISA in 39 children with JRA. The expressions of CD4(+)CD45RA(+)T and CD4(+)CD45RO(+)T in peripheral blood were detected by flow cytometry in 24 out of the 39 patients with JRA.
Surface-enhanced infrared absorption spectroscopy (SEIRAS) in the attenuated-total-reflection configuration (ATR-SEIRAS) has been applied for the first time to Ni electrodes. SEIRA-active Ni electrodes were obtained through initial chemical deposition of a 60-nm-thick Au underlayer on the reflecting plane of an ATR Si prism followed by potentiostatic electrodeposition of a 40-nm-thick Ni overlayer in a modified Watt's electrolyte. The Ni nanoparticle film thus obtained exhibited exceptionally enhanced IR absorption for the surface probe molecule CO while maintaining unipolar and normally directed bands.

Objective: To investigate microcosmic essentials of blood stasis syndrome (BSS) at the gene level.
Methods: One hundred and sixty patients with hypertension or diabetes mellitus, BSS or non-BSS, were strictly selected in disease-syndrome integrated mode according to diagnosis standard. The expression of CD62p gene and HSP70 gene in them were observed, compared and analyzed with blood biochemical indexes by experimental techniques, and compared to those in healthy subjects as control, for exploring the microcosmic mechanism and evolutive rules of BSS formation at the gene level.
Background: The aim of the present work was to study the potential effect of intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1) and neural cell adhesion molecule (NCAM) of immunoglobulin superfamily on the invasion and metastasis of salivary adenoid cystic carcinoma (SACC).
Method: The expressions of ICAM-1, VCAM-1 and NCAM of forty cases with SACC were examined by immunohistochemical methods using respective kits.
Results: A significant relation showed between the expression level and histological differentiation, expression of ICAM-1 and VCAM-1 in solid SACC was greatly increased compared with NCAM; SACC with metastatic lymph node or local recurrence displayed significant relationship to the up-regulation of ICAM-1 and VCAM-1, and to the down-regulation of NCAM.

Objective: To investigate the expression of matrix metalloproteinase-1 (MMP-1), tissue inhibitor of metalloproteinase-1 (TIMP-1) and transforming growth factor-beta 1 (TGF-beta 1) in human middle ear cholesteatoma.
