Diffraction-based automated crystal centering.

A fully automated procedure for detecting and centering protein crystals in the X-ray beam of a macromolecular crystallography beamline has been developed. A cryo-loop centering routine that analyzes video images with an edge detection algorithm is first used to determine the dimensions of the loop holding the sample; then low-dose X-rays are used to record diffraction images in a grid over the edge and face plane of the loop. A three-dimensional profile of the crystal based on the number of diffraction spots in each image is constructed.

Myofibroblasts and the progression of experimental glomerulonephritis.

We have studied the sequential morphological changes that took place in the kidneys of 8 rats with nephrotoxic serum nephritis (NTN). Rats underwent kidney biopsies at different time intervals (days 7, 15, 30, 90 and 120). The tissues were processed for light microscopy as well as immunohistochemistry for inflammatory cellular infiltrate as well as for the components of the extracellular matrix (ECM) and myofibroblasts (cells expressing alpha-smooth muscle actin, alpha-SMA).

Nutritional profile of continuous ambulatory peritoneal dialysis patients.

Undernutrition in dialysis patients contributes to their morbidity and mortality. This is a cross-sectional study of the nutritional status of 61 patients treated with continuous ambulatory peritoneal dialysis (CAPD). They were studied with emphasis on assessment of their nutritional intake, anthropometric measurements, and evaluation of biochemical parameters.

False aneurysm of the brachial artery: an unusual complication following accidental puncture by a patient on home haemodialysis.

A case is reported of false aneurysm of the brachial artery following accidental puncture during attempted cannulation of the median cubital vein for routine haemodialysis at home. The merits of early diagnosis and prompt surgical repair are discussed.

Continuous ambulatory peritoneal dialysis and haemodialysis in the elderly.

The clinical course and outcome of 58 patients receiving haemodialysis or continuous ambulatory peritoneal dialysis aged 65 years and above at the start of dialysis treatment was examined over a six-year period. Method of presentation and mode of treatment did not affect survival, but the presence of ischaemic heart disease or congestive cardiac failure was significantly associated with increased mortality. The actuarial two-year patient survival was 54 per cent.

Influence of selection for increased body weight on the incidence of leiomyomas and leiomyosarcomas in Japanese quail.

Leiomyomas and leiomyosarcomas were observed in adult laying hens from lines of Japanese quail selected solely (HW) or partly (HW-HP; HW-LP) for increased 4-wk BW and the corresponding randombred control (R1). No neoplasms were observed in a line (LW) selected for decreased 4-wk BW based on observations in one generation. Line R1 served as the base population for Lines HW and LW and was maintained without conscious selection for any trait, Lines HW-HP and HW-LP were sublines of Line HW in which the males were selected for increased 4-wk BW and the females were selected for high or low levels of plasma total phosphorus, respectively.

Testicular feminization in a cat.

Testicular feminization, caused by an inherited defect of the androgen receptor, was diagnosed in a domestic cat. Individuals affected with this syndrome are genetic males that have testes but fail to undergo masculinization because the internal and external genitalia cannot respond to androgens. The affected cat had the external appearance of a sexually normal female, but during surgery for ovariohysterectomy, only 2 abdominal gonads were found.

Lack of pathogenicity of five serotype 2 infectious bursal disease viruses in chickens.

The pathogenic potential of five strains of serotype 2 infectious bursal disease virus (IBDV) for specific-pathogen-free chickens was examined. There were no gross or microscopic lesions in the inoculated chickens. Bursa-to-body-weight ratios of IBDV-infected chickens were not significantly different from those of uninfected controls.

Pathogenesis of porcine enteric calicivirus-like virus in four-day-old gnotobiotic pigs.

Eighteen 4-day-old gnotobiotic pigs were orally inoculated with porcine enteric calicivirus-like virus (C strain). Seven additional gnotobiotic pigs served as noninoculated controls. Mild diarrhea developed in all inoculated pigs by postinoculation day (PID) 3 and persisted for 3 to 7 days.

Cholesterol embolisation: a lethal complication of vascular catheterisation.

Cholesterol embolisation after vascular catheterisation occurred in 5 patients. It tended to be associated with difficult manipulation of the catheter within a severely diseased aorta, and a common feature is leg pain with livedo reticularis despite palpable pulses. Confusion, renal failure, and death ultimately ensue.

Comparison of bone and muscle growth in turkey lines selected for increased body weight and increased shank width.

A subline (FL) of a long-term growth-selected line (F) of turkeys was established by mass selecting solely for increased shank width at the narrowest point (dew claw). A comparison was made of bone and muscle growth in Line FL (fourth generation of selection), Line F (17th generation of selection), and a randombred control (RBC2, the base population of F) at 8, 16, and 20 wk of age. We report three cases of subclavian vein obstruction following haemodialysis by a subclavian line. The obstruction only became apparent clinically following the formation of an arteriovenous fistula on the same side as the initial catheterisation. In each case, gross swelling of the arm followed creation of the fistula.

Chickens were inoculated with infectious bursal disease virus serotype I or serotype II to determine if their immune system can distinguish between the two serotypes. Chickens had neutralizing antibodies to only serotype I viruses following exposure to serotype I viruses, and chickens had antibodies to only serotype II viruses following exposure to serotype II viruses. No cross-reactions were observed between antisera prepared to each of these two serotypes using a cross-virus-neutralization assay.

A total of 128 isolates of Alcaligenes faecalis, from the respiratory tract of turkeys and chickens, were identified and divided into two types designated type I and type II. Type I isolates were pathogenic in poults, hemagglutinated guinea pig red blood cells (RBCs), and did not grow on minimal essential medium (MEM) agar, and most did not grow in 6.5% NaCl broth.

Porcine rotavirus-like virus (group B rotavirus): characterization and pathogenicity for gnotobiotic pigs.

A rotavirus-like virus (RVLV) was isolated from a diarrheic pig from an Ohio swine herd. This virus infected villous enterocytes throughout the small intestine of gnotobiotic pigs and induced an acute, transitory diarrhea. Complete virions were rarely observed in the intestinal contents of infected animals; the predominant particle detected by immune electron microscopy was a corelike particle 52 nm in diameter.

Impact of continuous ambulatory peritoneal dialysis on treatment of renal failure in patients aged over 60.

Thirty eight patients aged over 60 with end stage renal disease were treated by continuous ambulatory peritoneal dialysis for up to three years. Most of these patients, because of their age or coexisting diseases, had been considered to be unsuitable for haemodialysis by the criteria used before the advent of continuous ambulatory peritoneal dialysis in 1980. Actuarial patient survival at one and two years was 72% and 61% respectively, and only two patients were permanently transferred to haemodialysis.

Inherited XX sex reversal in the cocker spaniel dog.
