The primary structure of the messenger RNA coding for cytosolic phosphoenolpyruvate carboxykinase was determined by sequencing cDNA and genomic DNA and by primer extension of the mRNA. The molecule is 2624 nucleotides in length; this includes 143 nontranslated nucleotides at the 5' end and 615 nontranslated nucleotides at the 3' end. The 3' nontranslated sequence contains a 102-base pair region of alternating purine-pyrimidine nucleotides (the majority of which are UpG dinucleotides), several direct repeats and palindromic sequences, and 8 CpG dinucleotides.

To determine the long-term efficacy of insulin-pump therapy, we analyzed trends in glycosylated hemoglobin concentrations in 127 patients with Type I diabetes using insulin pumps for periods ranging from 13 to 47 months. In the first year of pump therapy the average glycosylated hemoglobin concentration improved in 83 per cent of the patients, as compared with the value before pump therapy. Although only 11 of the 127 subjects had normal glycosylated hemoglobin values before pump therapy, 33 had a normal average value during the first full year of pump use (P = 0.

In our wide experience of treating advanced breast carcinoma with chemotherapy, the combination of doxorubicin (DOX), vincristine (VCR), cyclophosphamide (CPM) and fluorouracil (FU) gave a complete plus partial response rate of over 60%, with 100% alopecia and frequent cardiac toxicity depending on total dose. After the EORTC Clinical Screening Group phase II trial we have conducted an "expected difference method" comparative phase II trial using the combination DOX, VCR, CPM, FU and the combination of MTX (10mg/m2), VCR, CPM and FU on a population of 50 breast carcinoma patients similar to those taking part in the first study. The reasons for similarity of action will be presented and discussed.

Procedures for the in vitro determination of the drug-induced inhibition of mammary and ovarian carcinoma cell growth were established. In monolayer cultures derived from advanced tumors, separation of epithelial carcinoma cells from concomitant cells of fibroblast-like or mesothelial appearance was achieved by differential trypsinization. The carcinoma cell character of the stock cultures was verified by chromosome analyses showing a high degree of aneuploidy for the epitheloid cell lines and euploidy for cells of apparently mesenchymal origin.

We determined the frequency of acute complications associated with insulin pump therapy in 161 insulin-dependent patients followed up for a total of 2,978 patient-months. Diabetes control improved substantively with pump therapy, but 42% of the patients experienced one or more acute complications while using insulin pumps. Infected infusion sites, ketoacidosis, and hypoglycemic coma occurred once in every 27, 78, and 175 patient-months, respectively.

The in vitro synthesis of murein from the precursors UDP-N-acetylglucosamine, L-alanine, D-glutamic acid and meso-diaminopimelic acid was performed with the aid of ether treated Escherichia coli. This synthesis was sensitive to representative inhibitors of early reactions in the cytoplasm as well as of late reactions in the membrane or the cell wall. The sensitivity was higher than in in vitro systems starting with UDP-N-acetylmuramic acid or UDP-N-acetylmuramyl-pentapeptide.

By using a translation assay in oocytes, a 17S RNA fraction coding for the interferon-induced (2'-5')oligo(A) synthetase was purified from human cells. A cDNA library was prepared by cloning in Escherichia coli plasmid pBR322 and screened by positive hybridization-translation in oocytes. Following locoregional treatment, patients were randomized into three groups: The first groups received no complementary treatment; the second group received adjuvant chemotherapy (vincristine, cyclophosphamide, and 5-fluorouracil once a month for 12 months); and the third group was treated by immunotherapy (150 mg BCG once a week for 1 year). Results of treating superficial cancerous nodules by hyperthermia, alone or combined with radiotherapy or chemotherapy, over a period of one year, are presented. Twenty-five patients with multiple cutaneous or subcutaneous metastatic tumors were treated by 1 to 3 MHz ultrasound hyperthermia or 434 MHz microwave hyperthermia. Good local tolerance was observed, even in regions previously treated with high doses of irradiation.

Incubation of rat red cells with liposomes containing radioactive glucosylceramide, together with unlabeled phosphatidylcholine and cholesterol, produced cells labeled with the glycolipid. Part of the bound cerebroside could be removed by further incubation with unlabeled liposomes. Both processes, uptake and release, were stimulated by adding a preparation obtained from bovine spleen.

Fourteen cases with a malignant dysembroma of the ovary were treated by resection, postoperative radiotherapy and/or chemotherapy. Eight of the 14 patients survived 2 years and 2 out of 7 are still alive 5 years later. The prognosis depends mainly on the initial stage of the disease.

A retrospective analysis of 95 treated nonseminomatous germinal testicular tumors is made. The primary treatment consisted of orchidectomy with high ligation of the cord. A histological study disclosed 53 embryonal cell carcinomas (56%), 21 chorioepithelionas (22%), 7 teratocarcinomas (7%) and 14 complex composite tumors which were predominantly dysembryomatous (15%).
