Previous studies have established that strain 68-1-derived rhesus cytomegalovirus (RhCMV) vectors expressing simian immunodeficiency virus (SIV) proteins (RhCMV/SIV) are able to elicit and maintain cellular immune responses that provide protection against mucosal challenge of highly pathogenic SIV in rhesus monkeys (RMs). However, these efficacious RhCMV/SIV vectors were replication and spread competent and therefore have the potential to cause disease in immunocompromised subjects. To develop a safer CMV-based vaccine for clinical use, we attenuated 68-1 RhCMV/SIV vectors by deletion of the Rh110 gene encoding the pp71 tegument protein (ΔRh110), allowing for suppression of lytic gene expression.

Enhancing safety of cytomegalovirus-based vaccine vectors by engaging host intrinsic immunity.

Rhesus cytomegalovirus (RhCMV)-based vaccines maintain effector memory T cell responses (T) that protect ~50% of rhesus monkeys (RMs) challenged with simian immunodeficiency virus (SIV). Because human CMV (HCMV) causes disease in immunodeficient subjects, clinical translation will depend upon attenuation strategies that reduce pathogenic potential without sacrificing CMV's unique immunological properties. We demonstrate that "intrinsic" immunity can be used to attenuate strain 68-1 RhCMV vectors without impairment of immunogenicity.

The terminology of identities between, outside and beyond the gender binary - A systematic review.

Recently, a multitude of terms have emerged, especially within North America and Western Europe, which describe identities that are not experienced within the culturally accepted binary structure of gender which prevails within those cultures. As yet, there is no clear single umbrella term to describe such identities and a mixture of words have been used in scholarly work to date. To explore the origins and track the emergence of newer terms and definitions for identities between, outside and beyond the gender binary, to outline current trends in descriptors within scholarly work and to suggest a term which is wide enough to encompass all identities.

Objectives Pregnant young women gain more weight than recommended by the National Academy of Medicine, increasing the likelihood of adverse maternal and fetal outcomes. The purpose of this study is to use online social media to understand beliefs and practices surrounding weight gain, diet and exercise during pregnancy among young women. Methods Facebook posts were mined from young women ages 16 to 24 during pregnancy who were consented from two Midwest primary care clinics serving low-income communities.

Meta-analysis in metastatic uveal melanoma to determine progression free and overall survival benchmarks: an international rare cancers initiative (IRCI) ocular melanoma study.

Background: Despite the completion of numerous phase II studies, a standard of care treatment has yet to be defined for metastatic uveal melanoma (mUM). To determine benchmarks of progression free survival (PFS) and overall survival (OS), we carried out a meta-analysis using individual patient level trial data.

Methods: Individual patient variables and survival outcomes were requested from 29 trials published from 2000 to 2016.

Objectives: Many older adults in long-term care (LTC) experience acute health crises but are at high risk of transfer distress and in-hospital morbidity and mortality. Residents often complete advance directives (ADs) regarding future care wishes, including directives for hospital transfers. This study aims to estimate the prevalence of, and adherence to, "no transfer to hospital" ADs in LTC, and to explore the circumstances leading to transfers against previously expressed directives.

A scalable database of organ doses for common diagnostic fluoroscopy examinations of children: procedures of current practice at the University of Florida.

Of all the medical imaging modalities that utilize ionizing radiation, fluoroscopy proves to be the most difficult to assess values of patient organ dose owing to the dynamic and patient-specific nature of the irradiation geometry and its associated x-ray beam characteristics. With the introduction of the radiation dose structured report (RDSR) in the mid-2000s, however, computational tools have been developed to extract patient and procedure-specific data for each irradiation event of the study, and when coupled to a computational phantom of the patient, values of skin and internal organ dose may be assessed. Unfortunately, many legacy and even current diagnostic fluoroscopy units do not have RDSR reporting capabilities, thus limiting these dosimetry reporting advances.

Subsequently, these genes have been shown to hold functional importance in human cellular biology, which can be exploited by tumors to drive the hallmarks of cancer. Due to the complex tertiary structure and unknown binding motifs of lncRNAs, there is a growing disparity between the number of lncRNAs identified and those that have been functionally characterized.

Previously undescribed thyroid-specific miRNA sequences in papillary thyroid carcinoma.

Papillary thyroid carcinoma (PTC) is the most common thyroid malignancy, wherein diagnostic limitations and lack of accurate prognostic factors are important clinical challenges. In this study, we report the discovery of 234 novel miRNAs in non-neoplastic thyroid and PTC samples, obtained from publicly available small RNA sequencing datasets (TCGA and GEO). These sequences were observed to display similar molecular features compared to currently annotated miRNAs.

Regional differences in where and how family medicine residents intend to practise: a cross-sectional survey analysis.

Background: Family medicine residents choose among a range of practice options as they enter the physician workforce. We describe the demographic and personal characteristics of Canadian family medicine residents and examine differences in the intentions of residents from Ontario, Quebec, Western Canada and Atlantic Canada at the completion of their training, in terms of practice comprehensiveness, organizational model, clinical domains, practice settings and populations served.

Methods: We analyzed national survey data collected by the College of Family Physicians of Canada and 16 university-based family medicine residency programs.

Fertility Preferences and Cognition: Religiosity and Experimental Effects of Decision Context on College Women.

Better models of culture and cognition may help researchers understand fertility and family formation. We examine cognition about fertility using an experimental survey design to investigate how fertility preferences of college women are affected by two prompts that bring to mind fertility-relevant factors: career aspirations and financial limitations. We test the effects of these prompts on fertility preferences and ask how effects vary with respondent religiosity, an aspect of social identity related to fertility preferences.

Direct ink writing of catalytically active UiO-66 polymer composites.

Metal-organic frameworks (MOFs) hold significant potential for use in gas storage, sensing and catalysis. To uncover this potential, MOF processing must develop in line with MOF materials. Here, direct ink writing-based 3D printing of UiO-66 MOF composites and their thermal treatment give mechanically stable yet highly porous composites effective for the catalytic breakdown of methyl-paraoxon, a simulant of highly toxic organophosphate nerve agents.

The purpose of this research was to evaluate an existing intervention aimed at empowering Indigenous youth, using a qualitative, community-based participatory research method. Background: The tumor microenvironment (TME) is a complex mixture of tumor epithelium, stroma and immune cells, and the immune component of the TME is highly prognostic for tumor progression and patient outcome. In lung cancer, anti-PD-1 therapy significantly improves patient survival through activation of T cell cytotoxicity against tumor cells. Direct contact between CD8+ T cells and target cells is necessary for CD8+ T cell activity, indicating that spatial organization of immune cells within the TME reflects a critical process in anti-tumor immunity.

The Implicit Activation Mechanism of Culture: A Survey Experiment on Associations with Childbearing.

This paper proposes a mechanism by which exposure to forms of culture "in the world" activates individuals' cognitive associations beneath conscious awareness, making certain behaviors more likely. A survey experiment illustrates part of the proposed mechanism, testing whether cues that make salient a shared cultural representation affect the activation of individuals' associations with childbearing. Drawing on cultural beliefs regarding the ostensible contradiction between close relations and monetary exchange, we expect that making one of these spheres salient would inhibit activation of associations with the other sphere.

Ambulatory management in low risk neutropenic sepsis - A plea for integrated acute cancer care.

Neutropenic Sepsis (NS) is a well recognised treatment complication, typically occurring 7-10 days following cancer cytotoxic chemotherapy. Colleagues in acute medicine will be only too familiar with the scenario of cancer patients that present with fever in the absence of localising signs and symptoms and with a very low yield from microbiological cultures. The incidence and mortality of NS are poorly defined and historically, management guidelines have often been developed in relative isolation from the broader subject of infection and sepsis care.

Imaging glial activation in patients with post-treatment Lyme disease symptoms: a pilot study using [C]DPA-713 PET.

The pathophysiology of post-treatment Lyme disease syndrome (PTLDS) may be linked to overactive immunity including aberrant activity of the brain's resident immune cells, microglia. Here we used [C]DPA-713 and positron emission tomography to quantify the 18 kDa translocator protein, a marker of activated microglia or reactive astrocytes, in the brains of patients with post-treatment Lyme disease symptoms of any duration compared to healthy controls. Genotyping for the TSPO rs6971 polymorphism was completed, and individuals with the rare, low affinity binding genotype were excluded.
