Introduction: The Brisbane Burn Scar Impact Profile (BBSIP) and the Patient and Observer Scar Assessment Scale (POSAS) are used in burn scar assessment to quantify patient health-related quality of life (HR-QoL). These questionnaires were developed using pen-and-paper delivery methods; however, there is a push towards electronic delivery of these questionnaires in both clinical practice and research. Equivalence testing is required to ensure that validity of these paper questionnaires is maintained electronically.

Study Question: How can we best achieve tissue segmentation and cell counting of multichannel-stained endometriosis sections to understand tissue composition?

Summary Answer: A combination of a machine learning-based tissue analysis software for tissue segmentation and a deep learning-based algorithm for segmentation-independent cell identification shows strong performance on the automated histological analysis of endometriosis sections.

What Is Known Already: Endometriosis is characterized by the complex interplay of various cell types and exhibits great variation between patients and endometriosis subtypes.

Study Design, Size, Duration: Endometriosis tissue samples of eight patients of different subtypes were obtained during surgery.

Similar to the mammalian hepatocytes, oenocytes accumulate fat during fasting, but it is unclear how they communicate with the fat body, the major lipid source. Using a modified protocol for prolonged starvation, we show that knockdown (KD) of the sole delta 9 desaturase, Desat1 (SCD in mammals), specifically in oenocytes leads to more saturated lipids in the hemolymph and reduced triacylglycerol (TAG) storage in the fat body. Additionally, oenocytes with KD exhibited an accumulation of lipoproteins and actin filaments at the cortex, which decreased lipoproteins in the hemolymph.

Article Synopsis
  • The study examined 33 severe COVID-19 patients who underwent a novel hemoperfusion therapy using the Seraph100 filter, aiming to reduce viral RNA levels and markers of inflammation.
  • Results showed that while the initial levels of SARS-CoV-2 RNA were generally low and did not significantly change post-treatment, five inflammatory protein analytes exhibited notable decreases, indicating some therapeutic effect on inflammation and endothelial damage.
  • The conclusion highlighted that Seraph 100 did not effectively lower viral RNA levels, but it did successfully reduce several inflammatory markers in the blood of these patients.

Article Synopsis
  • Research highlights the significant role of immune processes in the development of Alzheimer's disease, which is the leading cause of dementia.
  • Various studies indicate that both innate and adaptive immune responses contribute to the disease's pathology and are influenced by genetics and lifestyle factors.
  • New therapeutic approaches targeting neuroinflammation are being explored in clinical settings, offering potential treatment options for Alzheimer's patients.
