Background And Aims: The adipocyte-derived adiponectin (APN) has potent insulin-sensitizing and anti-inflammatory properties. The adipose tissue is known to be the main source for APN in the circulation, but sites and mechanisms which remove APN from blood are still unknown in humans.

Methods And Results: We reviewed APN data obtained in previous studies in which the inter-organ exchange of amino acids and cytokines was measured in our laboratory.

Nuclear laminar or inner nuclear membrane proteins, including lamin A, B1, and B2 and emerin, are involved in maintaining nuclear morphology. However, their expression patterns vary among tumors and remain incompletely understood. Endometrioid carcinoma (EC) exhibits mild nuclear atypia, although the underlying reasons have not been thoroughly explored.

Article Synopsis
  • No prior reports have compared section thickness (ST) between sliding microtomes (SM) and rotary microtomes (RM), leading to a study using two cooling conditions: ice pack (IP) and continuous cooling device (CCD).
  • Results showed that while ST was greater for SM under IP conditions, both microtomes had identical ST when using the CCD condition; thus, CCD was selected for further research.
  • The study found a significant difference in ST between tissue surfaces and paraffin surfaces in fish sausage blocks, but no difference in human kidney blocks, highlighting the importance of consistent ST and cooling methods for accurate quantitative analysis.

Article Synopsis
  • Emerin and lamins play a role in shaping the nucleus and may influence cell differentiation, particularly in lung adenocarcinoma.
  • A study of 82 lung adenocarcinoma cases found that the expression levels of lamin A, B1, B2, and emerin correlated with nuclear characteristics and differentiation grades, with lower levels in poorly differentiated tumors (G3).
  • Specifically, acinar adenocarcinomas with cribriform patterns showed significantly reduced expression of these proteins compared to other histological subtypes, indicating a potential link between protein levels and tumor aggressiveness.

Unlabelled: We measured section thickness (ST) after slicing using a film thickness meter and investigated the relationship between ST and the percent area of positive staining using computer-assisted image analysis.

Methods: Sections were prepared from a paraffin-only block and formalin-fixed paraffin-embedded (FFPE) blocks containing fish sausage and human liver specimens. The ST was compared between the sections prepared with cooling using an ice pack (IP) or a continuous cooling device (CCD) paired with a sliding microtome set at an ST of 4 µm.
