Background: Aging, tau pathology, and chronic inflammation in the brain play crucial roles in neuroinflammation, synaptic loss, neurodegeneration, and cognitive decline in tauopathies, such as Alzheimer's disease. However, the molecular mechanisms that trigger aberrant chronic inflammatory signaling in tauopathies are poorly understood.
Method: We utilized brain tissues from tauopathy patients and the tauopathy mouse models.
Paxalisib is a dual PI3K/mTOR inhibitor, being used in advanced cancer treatment. In this research, we report a validated LC-MS/MS method for quantifying paxalisib from mouse dried blood spot (DBS). We validated the method in-line with the FDA guidelines.

This resurgence is compounded by factors such as vaccine hesitancy, the impact of the COVID-19 pandemic on immunization rates, and international travel introducing new cases from endemic regions.
