Pyruvate metabolism defects lead to severe neuropathies such as the Leigh syndrome (LS) but the molecular mechanisms underlying neuronal cell death remain poorly understood. Here, we unravel a connection between pyruvate metabolism and the regulation of the epitranscriptome that plays an essential role during brain development. Using genetically engineered mouse model and primary neuronal cells, we identify the transcription factor E4F1 as a key coordinator of AcetylCoenzyme A (AcCoA) production by the pyruvate dehydrogenase complex (PDC) and its utilization as an essential co-factor by the Elongator complex to acetylate tRNAs at the wobble position uridine 34 (U).

Objectives: This study aimed to discuss teaching and assessment of advocacy skills in student pharmacists as called for in the American Association of Colleges of Pharmacy 2022 Curriculum Outcomes and Entrustable Professional Activities.

Findings: The American Association of Colleges of Pharmacy 2022 Curriculum Outcomes and Entrustable Professional Activities categorize advocacy as a skill that should be developed to advance patient care and the profession. This is a change from the 2013 Center for Advancement of Pharmacy Education outcomes, in which the focus was on advocating for patients.

Spinal muscular atrophy (SMA) is a pediatric genetic disorder characterized by the loss of spinal cord motor neurons (MNs). Although the mechanisms underlying MN loss are not clear, current data suggest that glial cells contribute to disease pathology. We have previously found that SMA astrocytes drive microglial activation and MN loss potentially through the upregulation of NF-κB-mediated pro-inflammatory cytokines.

Following regulatory pressure, the manufacture of long-chain per- and polyfluoroalkyl substances (PFAS) has been phased out, and alternatives such as short-chain homologs and ether-PFAS have replaced the bioaccumulative long-chain PFAS. However, data are lacking regarding the toxicokinetic (TK) properties of certain PFAS, particularly emergent substitutes for long-chain compounds. Additionally, the existing analytical methods used for TK studies measure a single compound or only a few simultaneously.

The behavior and removal of roxithromycin (ROX), oxytetracycline (OTC), chlortetracycline (CTC), and enrofloxacin (ENR) were investigated during the steady state of sludge anaerobic digestion (AD) in semi-continuous mode (37 °C). Sludge was spiked at realistic concentrations (50 μg/L of each antibiotic) and then used to feed the bioreactor for 80 days. Antibiotics were extracted from the substrate and digested sludge samples by accelerated solvent extraction (ASE).
