Article Synopsis
  • * After exposure to UV light, CS-B cells showed a delay in transcription progress and less dynamic chromatin compared to normal cells, revealing limitations in their response to DNA damage.
  • * The research highlights that despite these impairments, transcription initiation still occurs in CS-B fibroblasts shortly after UV exposure, indicating a complex role for CSB beyond its known function in DNA repair.

miRNAs constitute fine-tuners of gene expression and are implicated in a variety of diseases spanning from inflammation to cancer. miRNA expression is deregulated in rheumatoid arthritis (RA); however, their specific role in key arthritogenic cells such as the synovial fibroblast (SF) remains elusive. Previous studies have shown that expression is upregulated in RA SFs.

Cellular senescence constitutes a generally irreversible proliferation barrier, accompanied by macromolecular damage and metabolic rewiring. Several senescence types have been identified based on the initiating stimulus, such as replicative (RS), stress-induced (SIS) and oncogene-induced senescence (OIS). These senescence subtypes are heterogeneous and often develop subset-specific phenotypes.

Article Synopsis
  • Transcription is key for gene expression and maintaining genome integrity, especially after UV stress.
  • A study monitored how RNA Polymerase II assembles at gene promoters during recovery from UV damage, showing that recruitment happens faster shortly after exposure.
  • The research revealed different dynamics in gene assembly post-UV stress, which correlate with mutational patterns in melanoma tumors, highlighting the importance of active transcription during recovery.

The timing and location of writing and erasing of histone modifications determine gene expression programs and are tightly controlled processes. One such modification is the monoubiquitination of histone H2B (H2Bub), whose precise level during transcription elongation is dynamically regulated by the synergistic action of RNF20/40 ubiquitin-ligase and the de-ubiquitinase (DUB) of the ATXN7L3-containing DUB modules. Here, we characterize the dynamics of H2Bub in transcription and explore its role in perspective with the recently updated model of UV damage-induced transcription reorganization.
