Leucine-rich repeat kinase 2 (LRRK2) inhibition is a promising disease-modifying therapy for LRRK2-associated Parkinson's disease (L2PD) and idiopathic PD (iPD). However, pharmaco-dynamic readouts and progression biomarkers for clinical trials aiming for disease modification are insufficient since no endogenous marker reflecting enhanced kinase activity of the most common LRRK2 G2019S mutation has been reported yet in L2PD patients. Employing phospho-/proteomic analyses we assessed the impact that LRRK2 activating mutations had in peripheral blood mononuclear cells (PBMCs) from a LRRK2 clinical cohort from Spain (n=174).

Article Synopsis
  • - The Cannabis Refusal Self-Efficacy Questionnaire (CRSEQ) assesses people's confidence in resisting cannabis, consisting of 14 items that categorize into Emotional Relief Self-Efficacy, Opportunistic Self-Efficacy, and Social Facilitation, aiming to validate its use in the Chilean population.
  • - A study involving 1,275 Chilean individuals showed strong internal consistency and good fit for both 14-item and 19-item models, with the 14-item model demonstrating slightly superior fitting statistics, making it more applicable for various settings.
  • - The CRSEQ demonstrated convergent validity with cannabis use behaviors and was confirmed for demographic invariance, allowing meaningful comparisons across different sex and age groups, which is valuable for educational and

For cystic fibrosis patients, a lung-targeted gene therapy would significantly alleviate pulmonary complications associated with morbidity and mortality. However, mucus in the airways and cell entry pose huge delivery barriers for local gene therapy. Here, we used phage display technology to select for and identify mucus- and cell-penetrating peptides against primary human bronchial epithelial cells from cystic fibrosis patients cultured at the air-liquid interface.
