Objective: Elinzanetant is a selective neurokinin-1,3 receptor antagonist in development for the treatment of vasomotor symptoms (VMS) associated with menopause. The pivotal, double-blind, randomized, placebo-controlled phase 3 studies Overall Assessment of efficacy and Safety of elinzanetant In patients with vasomotor Symptoms (OASIS) 1 and 2 will assess the efficacy and safety of elinzanetant in women with VMS.

Methods: The OASIS 1 and 2 pivotal studies are designed in accordance with regulatory guidance.

Article Synopsis
  • Bacteria within human hosts face tradeoffs where mutations can be beneficial or harmful depending on the context.
  • Researchers found that a cystic fibrosis-related bacteria adapts by acquiring mutations that switch its traits, specifically changes in O-antigen expression.
  • The study indicates that whether O-antigen is beneficial or not depends on the infection site and duration, suggesting that continuous mutation during infection is more common than previously thought.

Cystic fibrosis (CF) is a genetic disorder caused by mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) that controls chloride current. A number of different CFTR transgenic mouse lines have been developed and subjected to both acute and chronic infection models. However, prior studies showed no substantial differences in bacterial clearance between CF and non-CF mice after single inoculations.

The genus encompasses multiple human pathogens, including potential bioterrorism agents, that are often extensively antibiotic resistant. The FixLJ pathway in is a two-component system that regulates virulence. Previous work showed that mutations arising during chronic infection confer increased virulence while decreasing the activity of the FixLJ pathway.
