An actin-binding protein, known as Calponin 3 (CNN3), modulates the remodeling of the actin cytoskeleton, a fundamental process for the maintenance of skeletal muscle homeostasis. Although the roles of CNN3 in actin remodeling have been established, its biological significance in myoblast differentiation remains largely unknown. This study investigated the functional significance of CNN3 in myogenic differentiation, along with its effects on actin remodeling and mechanosensitive signaling in C2C12 myoblasts.

Cortactin (CTTN) is an actin-binding protein regulating actin polymerization and stabilization, which are vital processes for maintaining skeletal muscle homeostasis. Despite the established function of CTTN in actin cytoskeletal dynamics, its role in the myogenic differentiation of progenitor cells remains largely unexplored. In this study, we investigated the role of CTTN in the myogenic differentiation of C2C12 myoblasts by analyzing its effects on actin cytoskeletal remodeling, myocardin-related transcription factor A (MRTFA) nuclear translocation, serum response factor (SRF) activation, expression of myogenic transcription factors, and myotube formation.

Local and systemic immunosuppression are prominent features of pancreatic cancer, rendering anti-tumor effector cells inactive and immunotherapeutic approaches ineffective. The spleen, an understudied point of antigen-presentation and T cell priming in humans, holds particular importance in pancreatic cancer due to its proximity to the developing tumor. As main effectors of antigen presentation, dendritic cells display antigens to lymphocytes, thereby bridging the innate and adaptive immune response.

Article Synopsis
  • Tumour cells rely on nutrients from their environment to survive, particularly in challenging conditions, and pancreatic cancer-associated fibroblasts (CAFs) play a key role in regulating this process by secreting acetate.* -
  • The enzyme acetyl-CoA synthetase short-chain family member 2 (ACSS2) helps cancer cells utilize this acetate, impacting their gene expression and enhancing their survival in acidic conditions by stabilizing the transcription factor SP1.* -
  • Disabling the ACSS2-SP1-SAT1 pathway in experimental mouse models leads to reduced tumour growth, emphasizing the importance of this metabolic interaction for cancer cell resilience under stress.*

Skeletal myogenesis is an intricate process involving the differentiation of progenitor cells into myofibers, which is regulated by actin cytoskeletal dynamics and myogenic transcription factors. Although recent studies have demonstrated the pivotal roles of actin-binding proteins (ABPs) as mechanosensors and signal transducers, the biological significance of WAVE2 (Wiskott-Aldrich syndrome protein family member 2), an ABP essential for actin polymerization, in myogenic differentiation of progenitor cells has not been investigated. Our study provides important insights into the regulatory roles played by WAVE2 in the myocardin-related transcription factor A (MRTFA)-serum response factor (SRF) signaling axis and differentiation of myoblasts.

The dynamic rearrangement of the actin cytoskeleton plays an essential role in myogenesis, which is regulated by diverse mechanisms, such as mechanotransduction, modulation of the Hippo signaling pathway, control of cell proliferation, and the influence of morphological changes. Despite the recognized importance of actin-binding protein Flightless-1 (FLII) during actin remodeling, the role played by FLII in the differentiation of myogenic progenitor cells has not been explored. Here, we investigated the roles of FLII in the proliferation and differentiation of myoblasts.

The prediction of algal blooms using traditional water quality indicators is expensive, labor-intensive, and time-consuming, making it challenging to meet the critical requirement of timely monitoring for prompt management. Using optical measures for forecasting algal blooms is a feasible and useful method to overcome these problems. This study explores the potential application of optical measures to enhance algal bloom prediction in terms of prediction accuracy and workload reduction, aided by machine learning (ML) models.

Cancers originating in the esophagus or esophagogastric junction constitute a major global health problem. Esophageal cancers are histologically classified as squamous cell carcinoma (SCC) or adenocarcinoma, which differ in their etiology, pathology, tumor location, therapeutics, and prognosis. In contrast to esophageal adenocarcinoma, which usually affects the lower esophagus, esophageal SCC is more likely to localize at or higher than the tracheal bifurcation.

Pancreatic cancer is difficult to resect due to its unique challenges, often leading to incomplete tumor resections. Fluorescence-guided surgery (FGS), also known as intraoperative molecular imaging and optical surgical navigation, is an intraoperative tool that can aid surgeons in complete tumor resection through an increased ability to detect the tumor. To target the tumor, FGS contrast agents rely on biomarkers aberrantly expressed in malignant tissue compared to normal tissue.

Progress in heterogeneous advanced oxidation processes (AOPs) is hampered by several issues including mass transfer limitation, limited diffusion of short-lived reactive oxygen species (ROS), aggregation of nanocatalysts, and loss of nanocatalysts to treated water. These issues have been addressed in recent studies by executing the heterogeneous AOPs in confinement, especially in the nanopores of catalytic membranes. Under nanoconfinement (preferably at the length of less than 25 nm), the oxidant-nanocatalyst interaction, ROS-micropollutant interaction and diffusion of ROS have been observed to significantly improve, which results in enhanced ROS yield and mass transfer, improved reaction kinetics and reduced matrix effect as compared to conventional heterogenous AOP configuration.

Surgery remains the only potentially curative treatment option for pancreatic cancer, but resections are made more difficult by infiltrative disease, proximity of critical vasculature, peritumoral inflammation, and dense stroma. Surgeons are limited to tactile and visual cues to differentiate cancerous tissue from normal tissue. Furthermore, translating preoperative images to the intraoperative setting poses additional challenges for tumor detection, and can result in undetected and unresected lesions.

Water pollution generated from intensive anthropogenic activities has emerged as a critical issue concerning ecosystem balance and livelihoods worldwide. Although optimizing wastewater treatment efficiency is widely regarded as the foremost step to minimize pollutants released into the environment, this widespread application has encountered two major problems: firstly, the significant variation of influent wastewater constituents; secondly, complex treatment processes within wastewater treatment plants (WWTPs). Based on the data collected hourly using real-time sensors in three different full-scale WWTPs (24 h × 365 days × 3 WWTPs × 10 wastewater parameters), this work introduced the potential application of Machine Learning (ML) to predict wastewater quality.

In recent years, immune therapy, notably immune checkpoint inhibitors (ICI), in conjunction with chemotherapy and surgery has demonstrated therapeutic activity for some tumor types. However, little is known about the optimal combination of immune therapy with standard of care therapies and approaches. In patients with gastrointestinal (GI) cancers, especially pancreatic ductal adenocarcinoma (PDAC), preoperative (neoadjuvant) chemotherapy has increased the number of patients who can undergo surgery and improved their responses.

Neoadjuvant systemic therapy is a preferred treatment approach for a number of tumor types due to many potential advantages over upfront surgery, including tumor downstaging, early treatment of micrometastatic disease, and providing an in vivo test of tumor biology. For colon cancer, current standard of care is upfront surgery followed by adjuvant systemic therapy in high-risk patients. Concerns about inaccurate radiological staging and tumor progression during preoperative treatment, as well the lack of randomized data demonstrating benefit, are among the reasons for the limited use of neoadjuvant therapy in this disease.
