We introduce DiffFit, a differentiable algorithm for fitting protein atomistic structures into an experimental reconstructed Cryo-Electron Microscopy (cryo-EM) volume map. In structural biology, this process is necessary to semi-automatically composite large mesoscale models of complex protein assemblies and complete cellular structures that are based on measured cryo-EM data. The current approaches require manual fitting in three dimensions to start, resulting in approximately aligned structures followed by an automated fine-tuning of the alignment.

N6-methyladenosine (m6A) methylation and abnormal immune responses are implicated in neurodegenerative diseases, yet their relationship in Alzheimer's disease (AD) remains unclear. We obtained AD datasets from GEO databases and used AD mouse and cell models, observing abnormal expression of m6A genes in the AD group, alongside disruptions in the immune microenvironment. Key m6A genes (YTHDF2, LRPPRC, and FTO) selected by machine learning were associated with the Notch pathway, with FTO and Notch1 displaying the strongest correlation.

Three types of solution treatment and aging were designed to reveal the α' decomposition and its effect on the mechanical properties of near-α Ti-80 alloy, as follows: solution at 970 °C then quenching (ST), ST + aging at 600 °C for 5 h (STA-1), and ST + aging 600 °C for 24 h (STA-2). The results show that the microstructures of the ST samples were mainly composed of equiaxed α and acicular α', with a large number of dislocations confirmed by the KAM results. After subsequent aging for 5 h, α' decomposed into acicular fine α and nano-β (intergranular β, intragranular β) in the STA-1 specimen, which obstructed dislocation motion during deformation, resulting in the STA-1 specimen exhibiting the most excellent yield strength (1012 MPa) and maintaining sufficient elongation (8.

The longest possible haplotype is chromosome haplotype that is a set of co-inherited alleles occurred on a single strand chromosome inherited from one parent. Standard whole-genome shotgun sequencing technologies are limited by the inability to independently study the haplotype of homologous chromosomes due to the short-reads sequencing strategy and disturbance of homologue chromosomes. Here, we investigated several types of chromosomal abnormalities by a dilution-based method to separate an intact copy of homologous chromosome from human metaphase cells, and then single chromosomes were independently amplified by whole-genome amplification methods, converted into barcoded sequencing libraries, and sequenced in multiplexed pools by Illumina sequencers.
