Article Synopsis
  • The JAK-STAT signaling pathway plays a key role in regulating gene transcription in response to cytokines and growth factors, and this study focused on characterizing STAT genes in lumpfish.
  • Various methods were used to analyze the evolutionary relationships and responses of the JAK-STAT pathway when exposed to bacteria and poly(I:C), a substance that mimics viral infections.
  • The results indicated that lumpfish possess several STAT genes, which are highly conserved, and showed significant upregulation of specific genes in response to bacterial and viral challenges, highlighting a foundation for future research and the potential for vaccination strategies against diseases affecting lumpfish.

The proinflammatory cytokines TNF-α and IL-6 are important mediators of inflammatory reactions and orchestrators of the immune system in vertebrate. In this study, we have identified TNF-α and IL-6 in lumpfish, molecular characterized them at mRNA and gene level, performed homology modelling and measured their gene expression in different tissues and upon in vitro stimulation. A comprehensive phylogenetic analysis of TNF-α teleost sequences give novel insight into the TNF -α biology.

We performed RNA sequencing, identified components of the immune system and mapped early immune responses of lumpfish (Cyclopterus lumpus) leukocytes following in vitro exposure to the pathogenic bacterium Vibrio anguillarum O1. This is the first characterization of immune molecules in lumpfish at the gene level. In silico analyses revealed that genes encoding proteins involved in pathogen recognition, cell signaling and cytokines in mammals and teleosts are conserved in lumpfish.

The present study was undertaken to compare the effects of intraperitoneally injected bacterial lipopolysaccharide (LPS) and yeast beta-glucan on lysozyme activity in Atlantic salmon, and to explore what organ(s) are responsible for the increase in plasma lysozyme activity induced by the compounds. The results indicated that LPS stimulates plasma lysozyme activity at least as efficiently as beta-glucan. The lysozyme gene was shown to be transcribed in head kidney, spleen, liver and intestine, and accumulation of transcript was demonstrated in response to both beta-glucan and LPS in all of these organs.

Serum amyloid A (A-SAA) has previously been reported to be an acute-phase protein in salmonids. Hepatocytes represent a major source of A-SAA in salmonids, but nothing is known about hepatocyte SAA synthesis in fish. In the present work, the expression of A-SAA transcripts in primary cultures of Atlantic salmon hepatocytes in response to macrophage derived cytokines, human recombinant cytokines and bacterial lipopolysaccharide (LPS) was studied by Northern blot analysis.
