Background: Low-temperature expression of recombinant proteins may be advantageous to support their proper folding and preserve bioactivity. The generation of expression vectors regulated under cold conditions can improve the expression of some target proteins that are difficult to express in different expression systems. The cspA encodes the major cold-shock protein from Escherichia coli (CspA).

Endophytes, microorganisms that live in the internal tissues and organs of the plants, are known to produce numerous bioactive compounds, including, at times, some phytochemicals of their host plant. For such reason, endophytes have been quoted as a potential source for discovering bioactive compounds, particularly, of medical interest. Currently, many non-communicable diseases are threatening global human health, noticeably: diabetes, neurodegenerative diseases, cancer, and other ailment related to chronic inflammation and ageing.

Article Synopsis
  • ATP citrate lyase (ACL) is crucial for producing acetyl-CoA, which is important in making fatty acids and carotenoids in yeasts; researchers isolated two genes and one cDNA for ACL in the yeast Phaffia rhodozyma.
  • The ACL genes have complex structures with long coding sequences, generating polypeptides that share high similarity with ACLs from other fungi and even humans.
  • This study is the first to characterize a recombinant ACL from a yeast, laying groundwork for future research on gene regulation, promoter sequences, and ACL's role in metabolic pathways.

Objective: To investigate the effect of hypoglycaemic, hypolipidemic, oxidative stress, insulin resistance and advanced glycation endproducts (AGEs) formation of hexane extract from Piper auritum.

Methods: The streptozotocin (STZ)-induced diabetic Wistar rats were treated with the hexane extract from Piper auritum leaves for 28 days and a set of biochemical parameters were studied including glucose level, total cholesterol, triglycerides, lipid peroxidation, liver and muscle glycogen, superoxide dismutase, catalase, glutathione peroxidase and glutathione reductase. The liver function was observed by determining glucose-6-phosphatase, glucokinase and hexokinase activities, and the effect of the hexane extract on insulin level and protein glycation.

ATP-citrate lyase (ACL) is the key cytoplasmic enzyme which supplies acetyl-CoA for fatty acids in oleaginous yeast. Although it has been suggested that fatty acid and carotenoid biosynthesis may have a common source of acetyl-CoA in Phaffia rhodozyma, the source for carotenoids is currently unknown. The purpose of this work was to analyze the development of ACL activity during batch cultures of P.
