Many protein bioconjugation strategies focus on the modification of lysine residues owing to the nucleophilicity of their amine side-chain, the generally high abundance of lysine residues on a protein's surface and the ability to form robustly stable amide-based bioconjugates. However, the plethora of solvent accessible lysine residues, which often have similar reactivity, is a key inherent issue when searching for regioselectivity and/or controlled loading of an entity. A relevant example is the modification of antibodies and/or antibody fragments, whose conjugates offer potential for a wide variety of applications.

Background: The prevalence and burden of atopic dermatitis (AD) are disproportionately high in individuals with skin of colour (SOC). Previous research shows that risk for xerosis and/or dyspigmentation is heightened in this population and may be more bothersome. However, there are no patient-reported instruments developed specifically for these disease sequelae in patients with SOC.

Objective: As COVID-19 restrictions are eased, there has been a lively debate on whether residency recruitment interviews should be held virtually or in-person. However, environmental impact has rarely been a focus of this debate and only by inference from limited survey data. In this study, we aimed to estimate the carbon emissions generated from air-travel versus in-person interviews in the general surgery residency recruitment.

Background: Pericardial fluid (PF) contains cells, proteins, and inflammatory mediators, such as cytokines, chemokines, growth factors, and matrix metalloproteinases. To date, we lack an adequate understanding of the inflammatory response that acute injury elicits in the pericardial space.

Objective: To characterize the inflammatory profile in the pericardial space acutely after ischemia/reperfusion.

Article Synopsis
  • - Dermatologists often encounter patients with inflammatory skin conditions and related psychological issues, but many lack formal training to address these concerns holistically.
  • - Body dysmorphic disorder (BDD), a psychiatric condition that leads to excessive worry about minor or nonexistent flaws in appearance, is prevalent among dermatology patients, particularly those with chronic skin conditions like acne.
  • - Adolescents are especially at risk for BDD due to high rates of acne and the influence of social media, which can amplify body image dissatisfaction; therefore, dermatologists should use screening tools and collaborate with mental health professionals for optimal patient care.

Introduction: The availability of quality primary health care (PHC) services in Nigeria is limited. The PHC system faces significant challenges and the improvement and expansion of PHC services is constrained by low government spending on health, especially on PHC. Out-of-pocket (OOP) expenditures dominate health spending in Nigeria and the reliance on OOP payments leads to financial burdens on the poorest and most vulnerable populations.

Purpose: The pandemic strained many usual methods of communication and training, forcing changes to selection processes for health professions programs. The purpose of this study was to determine, through interviewee feedback, the effectiveness of a virtual interview process when compared with a previous in-person interview process at one physician assistant (PA) program.

Methods: During the pandemic, the PA program adapted to a virtual holistic interview process to include interaction with current students and faculty.

Background: "Interview hoarding" is commonly used to refer to the concentration of interview offers among a small number of high-performing residency applicants. Theoretically, if the same applicants interview at every program, fewer rank lists will be submitted than open residency positions, leading to a "match crisis" with unfilled positions after the match. There are no published studies we are aware of that describe the observed distribution of residency interview offers among orthopaedic surgery applicants or the potential impact of "hoarding" on that distribution.

Importance Over-application and interviewing are believed to be widespread in residency recruitment. These may have increased during the 2021 virtual recruitment season. The increase does not correspond to an increase in available residency positions and likely results in more interviews with low probabilities of yielding matches.

Semiconducting polymer nanoparticles (SPNs) are explored for applications in cancer theranostics because of their high absorption coefficients, photostability, and biocompatibility. However, SPNs are susceptible to aggregation and protein fouling in physiological conditions, which can be detrimental for in vivo applications. Here, a method for achieving colloidally stable and low-fouling SPNs is described by grafting poly(ethylene glycol) (PEG) onto the backbone of the fluorescent semiconducting polymer, poly(9,9'-dioctylfluorene-5-fluoro-2,1,3-benzothiadiazole), in a simple one-step substitution reaction, postpolymerization.

In recent years, the uses of silver nanoparticles have increased, which lead to nanoparticles discharge into aquatic bodies which may, if not well controlled, have harmful effect on different organisms. This calls for the need to constantly evaluate the toxicity level of nanoparticles. In this study, green biosynthesized silver nanoparticles mediated by endophytic bacteria Cronobacter sakazakii (CS-AgNPs) were subjected to toxicity evaluation by brine shrimp lethality assay.

Article Synopsis
  • The Primary Mitochondrial Myopathy Symptom Assessment (PMMSA) is a 10-item tool designed to evaluate the severity of symptoms in patients with mitochondrial disease, assessing its psychometric properties based on data from a clinical trial.
  • Participants in the trial, mostly women with an average age of 45, filled out the PMMSA daily for about 14 weeks while testing elamipretide in a randomized, placebo-controlled study.
  • Results showed that while the PMMSA items did not form a single construct, four related to tiredness were grouped as a "general fatigue" score, which proved to be stable and correlated well with other health measures, with meaningful reductions being linked to symptom improvement.

One of the hallmarks of Alzheimer's disease (AD) are deposits of amyloid-beta (Aβ) protein in amyloid plaques in the brain. The Aβ peptide exists in several forms, including full-length Aβ1-42 and Aβ1-40 - and the N-truncated species, pyroglutamate Aβ3-42 and Aβ4-42, which appear to play a major role in neurodegeneration. We previously identified a murine antibody (TAP01), which binds specifically to soluble, non-plaque N-truncated Aβ species.
