Mutations in the ceramide transporter CERT (CERT1), which is involved in sphingolipid biosynthesis, are associated with intellectual disability, but the pathogenic mechanism remains obscure. Alpha-synuclein is a 15 kDa protein associated with neurodegenerative diseases such as Parkinson disease and multiple-system atrophy where pathological forms of alpha-synuclein aggregate and become neurotoxic. Here we describe the nonclinical program to support a first-in-human (FIH) single ascending dose (SAD) study for Lu AF82422, a human recombinant, anti-alpha-synuclein monoclonal antibody (mAb) in development for treatment of synucleinopathies. Alpha-synuclein is primarily expressed in brain, peripheral nerves and in blood cells.

Epstein-Barr Virus (EBV) has been associated with development of rheumatic connective tissue diseases like rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE) in genetically susceptible individuals. Diagnosis of RA and SLE relies on clinical criteria in combination with the presence of characteristic autoantibodies. In addition, antibodies to several EBV antigens have been shown to be elevated in patients with these diseases compared to healthy controls (HC).

Background: Accurate microbial diagnosis is crucial for effective management of prosthetic joint infections. Culturing of multiple intraoperative tissue samples has increased diagnostic accuracy, but new preparatory techniques and molecular methods hold promise of further improvement. The increased complexity of sampling is, however, a tough challenge for surgeons and assistants in the operation theatre, and therefore we devised and tested a new concept of pre-packed boxes with a complete assortment of swabs, vials and additional tools needed in the operating theatre for non-standard samples during a clinical study of prosthetic joint infections.

Mobilization and deposition in cows are different strategies of metabolism; hence, the aim was to study the possibility of reducing the crude protein (CP) supply during deposition to limit the use of protein supplements and minimize the environmental impact. A total of 61 Jersey and 107 Holstein cows were assigned to 4 mixed rations in a 2 × 2 factorial design with 2 concentrate to forage ratios (CFR) and 2 CP levels: high CFR (40:60) and recommended CP [16% of dry matter (DM); HCFR-RP], high CFR (40:60) and low CP (14% of DM; HCFR-LP), low CFR (30:70) and recommended CP (16% of DM; LCFR-RP), and low CFR (30:70) and low CP (14% of DM; LCFR-LP), where RP met the Danish recommendations. Cows were fed concentrate in an automatic milking unit.

The divalent metal complexes M(II){(SC6H4-2-PR2)-κ(2)S,P}2 (3-7, and 9-11) (M = Zn, Sn, or Pb; R = (i)Pr, (t)Bu, or Ph), the Sn(IV) complexes Sn{(SC6H4-2-PR2)-κ(2)-S,P}Ph2Cl (12 and 13) (R = (i)Pr and (t)Bu), and the ionic Sn(IV) complexes [Sn{(SC6H4-2-PR2)-κ(2)-S,P}Ph2][BPh4] (14 and 15) (R = (i)Pr and (t)Bu) have been prepared and characterized by multinuclear NMR spectroscopy and single crystal X-ray diffraction when suitable crystals were afforded. The Sn(II) and Pb(II) complexes with R = Ph, (i)Pr, or (t)Bu (5, 6, 9, and 10) demonstrated ligand "folding" hinging on the P,S vector-a behavior driven by the repulsions of the metal/phosphorus and metal/sulfur lone pairs and increased M-S sigma bonding strength. This phenomenon was examined by density functional theory (DFT) calculations for the compounds in both folded and unfolded states.

Aim: This paper presents the development and validation of a new assessment tool for qualitative research articles, which could assess trustworthiness of qualitative research articles as defined by Guba and at the same time aid clinicians in their assessment.

Background: There are more than 100 sets of proposals for quality criteria for qualitative research. However, we are not aware of an assessment tool that is validated and applicable, not only for researchers but also for clinicians with different levels of training and experience in reading research articles.

The vibrational dynamics of vacuum-isolated hydrogen-bonded complexes between water and the two simplest alcohols is characterized at low temperatures by Raman and FTIR spectroscopy. Conformational preferences during adaptive aggregation, relative donor/acceptor strengths, weak secondary hydrogen bonding, tunneling processes in acceptor lone pair switching, and thermodynamic anomalies are elucidated. The ground state tunneling splitting of the methanol-water dimer is predicted to be larger than 2.

The 16S rRNA genes of Pyrobaculum aerophilum and Pyrobaculum islandicum were amplified by the polymerase chain reaction, and the resulting products were sequenced directly. The two organisms are closely related by this measure (over 98% similar). However, they differ in that the (lone) 16S rRNA gene of Pyrobaculum aerophilum contains a 713-bp intron not seen in the corresponding gene of Pyrobaculum islandicum.
