The electrophoretic analysis of erythrocyte membrane proteins after cooling the reconstituted cells to moderately low temperatures (-30 degrees divided by -70 degrees C) has demonstrated that freezing at slow rates results in an increase in the oligomerization capability of spectrine, the main membrane skeleton protein and band 3 protein under the effect of diamide--a thioloxidizing agent. The data obtained testify to the fact that the membrane is a relatively cryoresistant cellular component at more rapid cooling rates in the presence of cryoprotective compounds.

The barrier properties of reconstituted and native erythrocyte membranes frozen to -30, -40 or -70 degrees C and stored for a month were studied. The release of markers, namely haemoglobin molecules, [14C] sucrose and K+ ions from cells and membrane structures was measured. The main changes in the barrier function of ghosts and cells have been found to be due to freeze-thawing rather than to storage conditions.
View Article and Find Full Text PDFThe freezing-thawing of erythrocytes under protection of 1,2-propane diol and glycerol was studied for its effect on the intracellular content of K+,Na+ and state of their passive and active transport. The low-temperature conservation of cells under the effect of penetrating cryoprotectors does not change essentially the content of univalent cations. A significant increase in the Na+ transport is observed with application of the cryoprotective medium on the base of 1,2-propane diol.

Based on the data concerning the content of ATP, 2,3-DFG, K+, and Na+ in depreserved red cells stored at 4 degrees C in resuspending media TsNIIGPK 8b and 8v it has been established that blood preserved under protection of 1,2-propanediol is comparable with that preserved under glycerin protection as regards its quality and is even better from the standpoint of some other indicators. Medium TsNIIGPK 8v may be used as resuspending medium for blood exposed to low temperature preservation under protection of 1,2-propanediol.
