Introduction: The triggering receptor expressed on myeloid cells 2 (TREM2) arginine-47-histidine (R47H) mutation is a significant risk for Alzheimer's disease (AD) with unclear mechanisms. Previous studies focused on microglial amyloid-β (Aβ) phagocytosis with less attention on the impact of TREM2 mutation on blood monocytes.
Methods: Bone marrow transplantation (BMT) models were used to assess the contribution of blood monocytes carrying TREM2 mutation to AD.
In the developing brain, immature synapses contain calcium-permeable AMPA glutamate receptors (AMPARs) that are subsequently replaced with GluA2-containing calcium-impermeable AMPARs as synapses stabilize and mature. Here, we show that this essential switch in AMPARs and neuronal synapse maturation is regulated by astrocytes. Using biochemical fractionation of astrocyte-secreted proteins and mass spectrometry, we identified that astrocyte-secreted chordin-like 1 (Chrdl1) is necessary and sufficient to induce mature GluA2-containing synapses to form.

As a rate-limiting enzyme in cholesterol biosynthesis, HMGCR undergoes rapid sterol-promoted degradation. In contrast, destruction of Insig-1 releases its inhibition on SREBP and stimulates the expression of lipogenic genes.

In the title compound, [Mn(C(10)H(10)N(2))(6)](ClO(4))(2), the Mn(II) ion, located on an inversion center, is coordinated by six N atoms from three pairs of symmetry-related 1-benzyl-1H-imidazole ligands in a distorted octa-hedral geometry. In the crystal, weak inter-molecular C-H⋯O hydrogen bonds link the complex cations and perchlorate anions.

The title compound, [Cu(2)(C(7)H(3)NO(4))(2)(C(12)H(12)N(6))(H(2)O)(2)]·H(2)O, displays a discrete dinuclear structure, in which the central Cu(II) atom is five-coordinated in a distorted square-based pyramidal coordination geometry and the flexible ligand 1,4-bis-(1,2,4-triazol-1-ylmeth-yl)benzene adopts a bis-monodentate bridging mode linking the Cu(II) atoms. It is further assembled by O-H⋯O hydrogen-bond inter-actions involving both the coordinated and uncoordinated water molecules. The latter exhibits half-occupancy.
