The global incidence of metabolic dysfunction-associated fatty liver disease (MAFLD) has risen sharply. This condition is strongly associated with the risk of cardiovascular disease (CVD), but how MAFLD affects the development and progression of CVD, particularly concerning vascular calcification, remains unclear. Herein, extracellular vesicles (EVs) are identified from steatotic hepatocytes as a trigger that accelerated the progression of both vascular intimal and medial calcification.

Article Synopsis
  • - Vascular calcification is a serious health issue linked to high rates of cardiovascular problems, and the role of antibiotics in affecting this condition through gut microbiota disruption is still uncertain.
  • - In studies using antibiotic treatments, researchers found that antibiotics worsened vascular calcification in mice by decreasing certain beneficial gut bacteria and short-chain fatty acids (SCFAs) in the body.
  • - Adding acetate was shown to help reduce calcium deposits in blood vessels and prevent the transformation of vascular smooth muscle cells into bone-like cells, with its effects possibly linked to specific metabolic processes involving glutathione.

Article Synopsis
  • Vascular calcification is a serious risk factor for cardiovascular problems in diabetes patients, but the underlying mechanisms are not fully understood.
  • A study found that levels of miR-32-5p were higher in type 2 diabetes patients with coronary artery calcification, correlating with the severity of calcification.
  • Researchers identified the CEBPB/miR-32-5p/GATA6 pathway as crucial in promoting vascular calcification, suggesting that targeting this pathway could help prevent and treat vascular issues in diabetes.

The study focuses on the control of nonlinear dynamic systems in the presence of parameter uncertainties, unmodeled dynamics, and external disturbances. The lumped perturbation is assumed to be bounded within a polynomial in the system state with the polynomial parameters and degrees unknown a priori such that it accommodates a quite wider range dynamic systems. Based on the studies in recent super-twisting algorithm designs and the idea from adaptive sliding mode control for nonlinear systems with uncertainties, we propose a novel adaptive super-twisting algorithm with exponential reaching law, or exponential super-twisting algorithm (ESTA), for the high-stability and acceptable accuracy control of the aimed nonlinear dynamics.

Obesity dramatically increases the risk of type 2 diabetes, fatty liver, hypertension, cardiovascular disease, and cancer, causing both declines in quality of life and life expectancy, which is a serious worldwide epidemic. At present, more and more patients with obesity are choosing drug therapy. However, given the high failure rate, high cost, and long design and testing process for discovering and developing new anti-obesity drugs, drug repurposing could be an innovative method and opportunity to broaden and improve pharmacological tools in this context.

Vascular calcification (VC) is highly correlated with cardiovascular disease morbidity and mortality, but anti-VC treatment remains an area to be tackled due to the ill-defined molecular mechanisms. Regardless of the type of VC, it does not depend on a single cell but involves multi-cells/organs to form a complex cellular communication network through the vascular microenvironment to participate in the occurrence and development of VC. Therefore, focusing only on the direct effect of pathological factors on vascular smooth muscle cells (VSMCs) tends to overlook the combined effect of other cells and VSMCs, including VSMCs-VSMCs, ECs-VMSCs, Macrophages-VSMCs, etc.
