Measurements were made of the pH of cytoplasm and lysosomes of cultured skin fibroblasts from healthy donors and from patients with lysosomal storage diseases (mannosidosis, Fabry's disease, and Krabbe's disease), and the effects of sucrose loading on normal fibroblasts were studied. The cytoplasmic pH of the pathological cells did not differ from control values, but the intralysosomal pH was significantly higher in sucrose-loaded normal fibroblasts and in cells from a patient with mannosidosis and from another with Fabry's disease. The change in pH observed accorded with an increase in size of the organelles, owing to accumulation of nonhydrolyzable compounds.

Changes in the size and dry mass of glioma C-6 intact cells in culture were investigated for 35-50 min at 5 min intervals by means of vital cytointerferometry. Rhythmic variations in the size, dry mass and protein concentration were thereby revealed in glioma cells. These variations fall into the category of circahoralian ones.

We compared the Feulgen hydrolysis curves (37 degrees C, 5 M HCl) of human blood lymphocytes fixed by the following four methods: 96% ethanol, 60 min at 20 degrees C; ethanol-acetone, 1:1, 120 min at 4 degrees C; ethanol-glacial acetic acid mixture (3:1), containing 2% of formaldehyde (EAF), 90 min at 20 degrees C; and ethanol-glacial acetic acid (3:1), 60 min followed by 5% chrome alum solution for 360 min at 20 degrees C. The best results were obtained with EAF-fixation, with respect to the highest amount of DNA-Schiff complex at the peak point of the curve, the longest "plateau" region and the smallest scattering of DNA-Schiff amount values along the "plateau". With other types of fixation the addition of polyethylene glycol (PEG) 6,000 to the hydrolysis solution resulted in modification of the shape of the hydrolysis curve so that it became nearly similar to the EAF-curve.

Scanning integrating visible cytospectrophotometry of gallocyanin-, amido black-, fast green- or ammoniacal silver-stained sections of various areas of the central nervous system of adult rats sacrificed at 4 h intervals has demonstrated the presence of circadian changes in RNA, total protein and total histone content as well as in arginine-rich to lysine-rich histone ratio in the neurons and in their perineuronal satellite neuroglia. The peak of the content per cell of the macromolecular components studied was in some cases elevated above their lowest level up to 1.7-fold.
