Donors (N = 25) were treated with AMD3100 at a dose of 240 mug/kg by subcutaneous injection, and leukapheresis was then initiated just 4 hours later. Two-thirds of the donors collected an allograft with a CD34(+) cell dose sufficient for transplantation after just one dose of AMD3100.

We screened the entire coding region of JAK1 by total exonic resequencing of bone marrow DNA samples from 94 patients with de novo AML. We identified 2 novel somatic mutations in highly conserved residues of the JAK1 gene (T478S, V623A), in 2 separate patients and confirmed these by resequencing germ line DNA samples from the same patients.

Background: The purpose of this study was to retrospectively evaluate the accuracy and reliability of a computer simulation system that predicts the results of an internal curvilinear mandibular distractor in the treatment of hemifacial microsomia and severe mandibular retrognathia.
Methods: A two-dimensional computer analysis and simulation system was developed based on cephalometric radiographs and patient photographs. The software was used to generate and superimpose digital templates, corresponding to the actual distractor and logarithmic spiral, onto the lateral cephalometric tracings.
Purpose Of Review: Severe congenital neutropenia is an inborn disorder of granulopoiesis. Mutations of ELA2, encoding neutrophil elastase, are present in approximately 50% of cases of severe congenital neutropenia and nearly all cases of cyclic neutropenia, a related disorder of granulopoiesis. Herein, we review recent studies of the molecular pathogenesis of severe congenital neutropenia, with an emphasis on those cases associated with mutations of ELA2.
