Mammalian pre-implantation development is a complex process involving sophisticated regulatory dynamics. WD repeat domain 36 (WDR36) is known to play a critical role in mouse early embryonic development, but its regulatory function in human embryogenesis is still elusive due to limited access to human embryos. The human pluripotent stem cell-derived blastocyst-like structure, termed a blastoid, offers an alternative means to study human development in a dish.

Background: Increasing evidence suggests that mesenchymal stem cells (MSCs) repair traumatized tissues primarily through paracrine secretion and differentiation into specific cell types. However, the role of epithelial differentiation of MSCs in cutaneous wound healing is unclear. This study aimed to investigate the epithelial differentiation potential of gingival tissue-derived MSCs (GMSCs) in epithelial cell growth medium and the mechanisms underlying their differentiation into an epithelial-like cell phenotype.

Article Synopsis
  • Distinguishing between nontuberculous mycobacterial (NTM) lung infections and pulmonary tuberculosis is difficult due to their similar symptoms, leading to frequent misdiagnoses and delays in proper treatment.
  • This study investigated the effectiveness of smear-positive and Xpert assay negative results as early indicators of NTM infection in areas with high TB prevalence, finding that a significant number of cases showed this pattern.
  • The research concluded that using smear-positive/Xpert-negative outcomes can reliably signal the presence of NTM, potentially helping about 25% of affected patients with a quick screening method available from the first hospital visit.

COPD poses a significant global public health challenge, primarily characterised by irreversible airflow restriction and persistent respiratory symptoms. The hallmark pathology of COPD includes sustained airway inflammation and the eventual destruction of lung tissue structure. While multiple risk factors are implicated in the disease's progression, the underlying mechanisms remain largely elusive.

Lithium-sulfur batteries (LSB) show excellent potential as future energy storage devices with high energy density, but their slow redox kinetics and the shuttle effect seriously hinder their commercial application. Herein, a 0D@2D composite was obtained by anchoring polar nano-TiO onto a 2D layered g-CN surface in situ, and a functional separator was prepared using multi-walled carbon nanotubes as a conductive substrate. Due to their long-range conductivity, multi-walled carbon nanotubes make up for the low conductivity of TiO@g-CN to some extent.

The nuclear factor κB (NF-κB) pathway plays essential roles in innate and adaptive immunity, but little is known how NF-κB signaling is compartmentalized and spatiotemporally activated in the cytoplasm. Here, we show that the lipogenesis signal cascade Scap-SREBP1-S1P/S2P orchestrates the homeostasis and spatiotemporal activation of NF-κB. SREBP cleavage-activating protein (Scap) and sterol regulatory element-binding protein 1 (SREBP1) form a super complex with inhibitors of NF-κB α (IκBα) to associate NF-κB close to the endoplasmic reticulum (ER).
