ZGDHu-1 induces G₂/M phase arrest and apoptosis in Kasumi-1 cells.

The present study examined the effects of N,N'‑di‑(m‑methylphenyi)‑3, 6‑dimethyl‑1, 4‑dihydro‑1,2,4,5‑tetrazine‑1,4‑dicarboamide (ZGDHu‑1), a novel oxazine derivative, in Kasumi‑1 cells. Following incubation with various concentrations of ZGDHu‑1, fluorescence‑activated cell sorting (FACS) was used in order to detect changes in mitochondrial membrane permeability in Kasumi‑1 cells. Western blot analysis was performed in order to analyze the expression of nuclear factor‑κB, inhibitor of κB and AML1/ETO.

[Macrocalin A induces apoptosis of multiple myeloma U266 cells through inhibiting the proteasome].

This study was purposed to investigate the inhibitory effect of macrocalin A (MA) on proteasome of multiple myeloma U266 cells in vitro and molecular mechanism of MA-inducing apoptosis. U266 cells in vitro were incubated with different concentrations (2, 4, 8 µg/mL) of MA, the Hochest staining and Annexin-V/PI double staining were used to detect the apoptosis of U266 cells. The expressions of protein β1, β1i, β2, β2i, β5, β5i, ubiquitous, 19S subunit S6', and BAD,BCL-2, FAS, FAS-L,MAPK, PARP, Pro-caspase 3, cleaved-caspase 3 were detected by Western blot technique.
