Taking advantage of the drop-on-demand capabilities of inkjet printing, the first example of a single tissue being used as a substrate for preparing combinatorial arrays of different matrix-assisted laser desorption/ionization (MALDI) matrices in multiple concentrations on a single chip is reported. By varying the number of droplets per spot that were printed, a gradient array of different amounts of matrix material could be printed on a single chip, while the selection of matrices could be adjusted by switching different matrix materials. The result was a two-dimensional array of multiple matrices on a single tissue slice, which could be analyzed microscopically and by MALDI to elucidate which combination of matrix and printing conditions offered the best resolution in terms of spot-to-spot distance and signal-to-noise ratios for proteins in the recorded MS spectra.

In recent years, matrix-assisted laser desorption/ionization (MALDI)-imaging mass spectrometry has become a mature technology, allowing for reproducible high-resolution measurements to localize proteins and smaller molecules. However, despite this impressive technological advance, only a few papers have been published concerned with computational methods for MALDI-imaging data. We address this issue proposing a new procedure for spatial segmentation of MALDI-imaging data sets.

Carcinoma tissue consists of not only tumor cells but also fibroblasts, endothelial cells or vascular structures, and inflammatory cells forming the supportive tumor stroma. Therefore, the spatial distribution of proteins that promote growth and proliferation in these complex functional units is of high interest. Matrix-assisted laser desorption/ionization imaging mass spectrometry is a newly developed technique that generates spatially resolved profiles of protein signals directly from thin tissue sections.

Annexin A5 is a Ca(2+)-binding protein which is involved in membrane organization and dynamics. As recent data suggest a role of annexin A5 in cancer we aimed to gain more insight into the biological function of endogenous annexin A5 and assessed its possible influence on proliferation and invasion capacity. We downregulated annexin A5 by RNA interference in HaCaT keratinocytes, squamous carcinoma cell line A431 as well as in a primary cell culture of a human oral carcinoma.
