Metallo-organic cages possess flexibility comparable to that of biological receptors and can alter their conformations to better accommodate guest species due to the dynamic reversibility of the coordination bond. Induced fit is widely accepted involving conformation change of the host, while few definitive examples are related to conformation selection. Herein, we report the generation of metallo-organic cube Cd L with two coexisting conformations, which have been fully confirmed by NMR, ESI-MS and single-crystal X-ray diffraction analysis.

The chirality of metal-organic cages holds enormous potential for novel applications in diverse fields, while it is relatively rare to employ such asymmetric units for the construction of noncentrosymmetric materials. Herein, by self-assembling the 4,4',4''-nitrilotribenzoic acid (HNBA) with bis(cyclopentadienyl)-zirconium dichloride (CpZrCl, Cp=η-CH) in different solvent conditions, we have obtained three hierarchical packing modes of metallo-tetrahedra with distinct spatial symmetry groups (designated as Zr-α, Zr-β, and Zr-γ). Among them, Zr-α employs a simple cubic arrangement and is a common centrosymmetric superstructure, which consists of a pair of equimolar metallo-tetrahedra enantiomers in its unit cell.

Article Synopsis
  • Renal fibrosis is a key feature of chronic kidney disease, and Smad3 is a crucial factor in the pro-fibrosis signaling pathway activated by TGF-β, which promotes fibrosis in the kidneys.* -
  • The study found that Biochanin A (BCA), a natural compound, can significantly inhibit TGF-β signaling and reduce fibrotic gene expression, specifically targeting Smad3 while leaving another factor, Smad2, unaffected.* -
  • Research revealed that Klf6, a transcription factor that promotes Smad3 expression, is downregulated by BCA, which prevents the fibrotic effects mediated by TGF-β in renal fibrosis models.*

Renal fibrosis represents a pivotal characteristic of chronic kidney disease (CKD), for which effective interventions are currently lacking. The Src kinase activates the phosphatidylinositol-3 kinases (PI3K)/Akt1 pathway to promote renal fibrosis, casting a promising target for anti-fibrosis treatment. Chaihuang-Yishen formula (CHYS), a traditional Chinese medicinal prescription, has a validated efficacy in the treatment of CKD, however, with the underlying mechanism unresolved.

This study investigated the protective effect of ginsenoside Rg_1(GRg_1) on oxygen and glucose deprivation/reoxygenation(OGD/R)-injured rat adrenal pheochromocytoma(PC12) cells and whether the underlying mechanism was related to the regulation of inositol-requiring enzyme 1(IRE1)-c-Jun N-terminal kinase(JNK)-C/EBP homologous protein(CHOP) signaling pathway. An OGD/R model was established in PC12 cells, and PC12 cells were randomly classified into control, model, OGD/R+GRg_1(0.1, 1, 10 μmol·L~(-1)), OGD/R+GRg_1+rapamycin(autophagy agonist), OGD/R+GRg_1+3-methyladenine(3-MA,autophagy inhibitor), OGD/R+GRg_1+tunicamycin(endoplasmic reticulum stress agonist), OGD/R+GRg_1+4-phenylbutyric acid(4-PBA, endoplasmic reticulum stress inhibitor), and OGD/R+GRg_1+3,5-dibromosalicylaldehyde(DBSA, IRE1 inhibitor) groups.

Unlabelled: Delayed graft function (DGF) is one of the key post-operative challenges for a subset of kidney transplantation (KTx) patients. Graft survival is significantly lower in recipients who have experienced DGF than in those who have not. Assessing the risk of chronic graft injury, predicting graft rejection, providing personalized treatment, and improving graft survival are major strategies for predictive, preventive, and personalized medicine (PPPM/3PM) to promote the development of transplant medicine.

Hypoxia inducible factor 1α (HIF1α) is abnormally overexpressed in t(8;21) acute myeloid leukemia (AML) and functions as an oncogene through transactivating DNA methyltransferase 3 alpha leading to DNA hypermethylation. However, it remains unclear whether HIF1α influences RNA N-methyladenosine (mA) methyltransferases. Here, we show that HIF1α promotes the expression of Wilms tumor 1-associated protein (WTAP), a main component of the mA methyltransferase complex, markedly alters the transcriptome-wide mA distribution and enhances cell proliferation in t(8;21) AML.
