Background: Soft tissue sarcomas of the trunk wall (STS-TW) are usually studied together with soft tissue sarcomas of other locations. We report a study on STS-TW forming part of the French Sarcoma Group database.

Patients And Methods: Three hundred and forty-three adults were included.

Simultaneous characterization of optical and rheological properties of carotid arteries via bimodal spectroscopy: experimental and simulation results.

This study aimed at identifying potential correlations between rheological and optical properties of carotid artery rings before and after cryopreservation at different mechanical deformations using experimental and simulation results. Therefore, a uniaxial mechanical test bench was coupled to fibered optical spectroscopes measuring 410 nm excited autofluorescence and 650-850 nm elastically backscattered intensity spectra. Furthermore, we developed a statistical simulation program of light transport and fluorescence adapted to our specific experimental configuration.

Preventing bladder tumor implantation with photodynamic therapy in a rat model mimicking post-fluorescence guided transurethral resection.

Purpose: Fluorescence guided transurethral resection has gained acknowledgment from the urological community and it is progressively becoming more applied. It has been shown to decrease the recurrence rate of nonmuscle invasive bladder cancer due to incomplete resection due to lack of visualization. The implantation of viable tumor cells seeded during transurethral resection is another reason for recurrence.

Effect of a community-based Argentine tango dance program on functional balance and confidence in older adults.

Tango-dancing and walking programs are compared in nondemented seniors at risk for falls. Fallers (N = 30) age 62-91 were randomly assigned to a 10-wk (40 hr, 2 hr 2x/wk) tango class or walk group. The Activities-specific Balance Confidence (ABC) scale, sit-to-stand scores, and normal and fast walk were measured pre-, post-, and 1 month postintervention.

Damaged DNA binding protein 2 plays a role in breast cancer cell growth.

The Damaged DNA binding protein 2 (DDB2), is involved in nucleotide excision repair as well as in other biological processes in normal cells, including transcription and cell cycle regulation. Loss of DDB2 function may be related to tumor susceptibility. However, hypothesis of this study was that DDB2 could play a role in breast cancer cell growth, resulting in its well known interaction with the proliferative marker E2F1 in breast neoplasia.

Data concerning the fine structure of the 12q13-15 amplicon which contains MDM2 and CDK4 in well-differentiated and dedifferentiated liposarcomas (WDLPS/DDLPS) are scarce. We investigated a series of 38 WDLPS/DDLPS using fluorescence in situ hybridization analysis with 17 probes encompassing the 12q13-15 region. In addition, using quantitative RT-PCR we studied the expression of MDM2, CDK4, DDIT3 (CHOP/GADD153), DYRK2, HMGA2, TSPAN31 and YEATS4 (GAS41) in 11 cases.

Translocation-positive low-grade fibromyxoid sarcoma: clinicopathologic and molecular analysis of a series expanding the morphologic spectrum and suggesting potential relationship to sclerosing epithelioid fibrosarcoma: a study from the French Sarcoma Group.

Low-grade fibromyxoid sarcomas (LGFMS) bear either the t(7,16) (q32-34;p11) or t(11,16) (p11;p11) translocations, resulting in FUS-CREB3L2 or FUS-CREB3L1 fusions, respectively. Heretofore, fusion transcripts were mainly detected in frozen tissues, using reverse transcription-polymerase chain reaction. In this study, we aimed to develop a reliable method to detect these in paraffin-embedded tissues, and to examine the clinicopathologic characteristics of a series of translocation-positive LGFMS.

Vascular proliferations of the skin after radiation therapy for breast cancer: clinicopathologic analysis of a series in favor of a benign process: a study from the French Sarcoma Group.

Background: Cutaneous vascular proliferations that occur in the field of prior radiotherapy include angiosarcoma and small, cutaneous lesions with a pseudosarcomatous pattern that previously were reported as atypical vascular lesions or benign lymphangiomatous papules.

Methods: The objective of this study was to investigate the clinicopathologic features and outcomes of 56 radiation-induced vascular proliferations that occurred in 36 patients who received previous treatment for breast carcinoma. Data from all patients were retrieved from the files of the French Sarcoma Group.

Task-oriented intervention in chronic stroke: changes in clinical and laboratory measures of balance and mobility.

Objective: The objective of this study was to evaluate whether, after a task-oriented exercise program, the changes in clinical measures of balance and mobility were paralleled by changes in biomechanical parameters in subjects with chronic stroke.

Design: Ten stroke subjects took part in an 8-wk exercise program aimed at improving balance and mobility through various functional tasks. Subjects were evaluated before and after the exercise intervention.

A novel orthotopic bladder tumor model with predictable localization of a solitary tumor.

An ideal bladder tumor model consists in an orthotopic solitary tumor with a well-defined localization and stage, as well as unaltered normal mucosa. None of the existing models covers all these requirements. We have created a new model, suitable for diagnostic and therapeutic purposes.

Functional characterization and subcellular localization of the three malate dehydrogenase isozymes in Leishmania spp.

As part of a study on the malate dehydrogenase isozymes (MDHs) from Trypanosomatids, three different fractions with MDH activity were obtained from crude extracts of Leishmania mexicana promastigotes combining two different chromatographic steps. Gel filtration chromatography in native conditions showed that most of the MDH activity present in the crude extracts eluted in a single peak, which corresponded to a lower apparent molecular mass ( congruent with 57kDa) than the value expected for typical MDHs. To further characterize the leishmanial isozymes, three putative MDH genes, presumably corresponding to the mitochondrial, glycosomal and cytosolic isoforms were amplified by PCR, cloned into bacterial expression vectors, and the recombinant enzymes purified.

Postural adaptation to walking on inclined surfaces: II. Strategies following spinal cord injury.

Objective: To investigate the postural adaptations to inclined walking in spinal cord injured (SCI) subjects.

Methods: Eight subjects with an incomplete spinal cord injury and eight age- and sex-matched healthy control subjects walked on a treadmill at five different grades (from -10 to 10%) without any assistance. The movements of the trunk and pelvis were recorded with four high-resolution cameras.

Eradication of p53-mutated head and neck squamous cell carcinoma xenografts using nonviral p53 gene therapy and photochemical internalization.

Photochemical internalization (PCI) technology has been used for PEI-mediated p53 gene transfer in mice bearing head and neck squamous cell carcinoma (HNSCC) xenografts. Using luciferase as a reporter gene, PCI led to a 20-fold increase in transgene expression 48 h after transfection and sustained transgene expression for 7 days. Therefore, iterative p53 gene transfer was performed by means of a weekly single injection of PEIGlu4/p53 complexes alone or with PCI for 5 (group A) or 7 (group B) weeks.

Vastus medialis: anatomical and functional considerations and implications based upon human and cadaveric studies.

Objective: To provide an electrophysiological and functional description of the vastus medialis (VM) and contrast it to an anatomical description.

Methods: Motor points of all superficial portions of the quadriceps were identified on the dominant side of 8 human subjects and electrically stimulated to achieve a light contraction to trace and measure the orientation of the fibers. Electromyography of the VM was then recorded over 2 motor points during isometric and isokinetic maximum knee extensions.

Balance training following stroke: effects of task-oriented exercises with and without altered sensory input.

The purpose of this study was to compare the effects of a task-oriented exercise program with and without altered sensory input on postural stability in subjects with stroke. Sixteen hemiparetic subjects, at least 6 months post-stroke, were randomly assigned to the experimental or control group, and participated in an 8-week task-oriented exercise program focusing on balance and mobility exercises. Exercises were performed under normal conditions by the control group, and under conditions of vision and surface manipulation by the experimental group.

Results of preoperative lymphoscintigraphy for breast cancer are predictive of identification of axillary sentinel lymph nodes.

The aim of this study was to identify the variables associated with successful peroperative sentinel lymph node (SLN) localization. We studied 201 patients with T1, T2, N0 invasive breast cancer who underwent a SLN procedure from 1999 to 2003. Of these 201 patients, 55 underwent peritumoral and 146 underwent periareolar radioisotope injection before the blue dye injection.

Endoscopic confocal fluorescence microscopy of normal and tumor bearing rat bladder.

Purpose: We evaluated the possibility of performing endoscopic fiber-optic confocal microscopy in a rat bladder model and we distinguished different cell types.

Material And Methods: Rhodamine 123 (Molecular Probes, Eugene, Oregon) (100 microM) was instilled for 30 minutes in 5 tumor bearing rat bladders (AY27). Five normal rats served as controls.

Prenatal diagnosis of recessive congenital methaemoglobinaemia type II: novel mutation in the NADH-cytochrome b5 reductase gene leading to stop codon read-through.

A case of type II recessive congenital methaemoglobinaemia (RCM) observed in a Lebanese subject with a novel mutation in NADH-cytochrome b5 reductase gene is described. A homozygous mutation CAC to AA identified at Thr 295 with an out-of-frame 1-bp deletion leads to a frameshift with translational read-through of the natural stop codon. The molecular mechanism is demonstrated by an in vitro translation study.

Exercise training to improve motor performance in chronic stroke: effects of a community-based exercise program.

The purpose of this study was to investigate the feasibility and efficacy of an exercise program offered by a community organization on motor performance in individuals with chronic stroke. Twenty hemiparetic individuals at least 6 months after a stroke and discharged from all rehabilitation services enrolled in the study. They took part in an 8-week group program (two times per week) aimed at improving balance, mobility, coordination, walking endurance and strength on the hemiparetic side through various functional exercises.
