Long-term potentiation in hippocampal neurons has stages that correspond to the stages of learning and memory. Early-phase (10-30 min) potentiation is accompanied by rapid increases in clusters or puncta of presynaptic and postsynaptic proteins, which depend on actin polymerization but not on protein synthesis. We have now examined changes in pre- and postsynaptic puncta and structures during glutamate-induced late-phase (3 hr) potentiation in cultured hippocampal neurons.

Dendritic spines are small protrusions from dendritic shafts that contain the postsynaptic sites of glutamatergic synapses in the brain. Spines undergo dramatic activity-dependent structural changes that are particularly prominent during neuronal development. Although changes in spine shape or number have been proposed to contribute to forms of synaptic plasticity that underlie learning and memory, the extent to which spines remain plastic in the adult brain is unclear.

Although Ca2+ release from internal stores has been proposed to be important for the induction of long-term synaptic plasticity, the importance of Ca2+ stores localized in presynaptic terminals remains unclear. Here, we have selectively applied pharmacological antagonists to either the presynaptic or postsynaptic cell in paired whole-cell recordings from hippocampal CA3 pyramidal neurons in slice culture. We demonstrate directly the necessary role of presynaptic, but not postsynaptic, ryanodine-sensitive Ca2+ stores in the induction of NMDA receptor (NMDAR)-dependent long-term depression (LTD).

Dopamine input to the striatum is required for voluntary motor movement, behavioral reinforcement, and responses to drugs of abuse. It is speculated that these functions are dependent on either excitatory or inhibitory modulation of corticostriatal synapses onto medium spiny neurons (MSNs). While dopamine modulates MSN excitability, a direct presynaptic effect on the corticostriatal input has not been clearly demonstrated.

The site of modification of synaptic transmission during long-term plasticity in the mammalian hippocampus remains controversial. Here we used a fluorescent marker of presynaptic activity, FM 1-43, to directly image presynaptic function during metabotropic glutamate receptor-dependent long-term depression (mGluR-LTD) at CA3-CA1 excitatory synapses in acute hippocampal slices. We found a significant decrease in the rate of FM 1-43 release in response to synaptic stimulation following induction of mGluR-LTD, providing direct evidence for altered presynaptic function.
