Purpose Low-dose total skin electron beam therapy (LD-TSEBT) has recently gained popularity in treating mycosis fungoides (MF) due to its reduced toxicity and favorable response rates. Combining accelerated LD-TSEBT with the modified Stanford technique (mST), a condensed cycling approach, offers a promising and convenient option. However, in vivo dosimetry data confirming the effectiveness of this approach is limited.

Antibody CAP256-VRC26.25 targets the second hypervariable region (V2) at the apex of the HIV envelope (Env) trimer with extraordinary neutralization potency, although less than optimal breadth. To improve breadth, we linked the light chain of CAP256V2LS, an optimized version of CAP256-VRC26.

Article Synopsis
  • *Production Challenges: Issues included sticking to pipette tips during dilution and proteolytic cleavage of its heavy chain, which was resolved by a targeted amino acid change to prevent this cleavage.
  • *Final Outcome: The modified version, CAP256V2LS, maintains its potent neutralization capabilities, improved stability in the body, and no signs of autoreactivity, making it a promising candidate for clinical development.
