CD4 effector lymphocytes (T) are traditionally classified by the cytokines they produce. To determine the states that T cells actually adopt in frontline tissues in vivo, we applied single-cell transcriptome and chromatin analyses to colonic T cells in germ-free or conventional mice or in mice after challenge with a range of phenotypically biasing microbes. Unexpected subsets were marked by the expression of the interferon (IFN) signature or myeloid-specific transcripts, but transcriptome or chromatin structure could not resolve discrete clusters fitting classic helper T cell (T) subsets.

In this study, quantitative model of two-photon excitation and fluorescence spectra of carbon monoxide based on up-to-date spectroscopic constants collected during an extensive literature survey was developed. This semi-classical model takes into account Hönl-London factors, quenching effects (collisional broadening and shift), ionization and stark effect (broadening and shift), whereas predissociation is neglected. It was specifically developed to first reproduce with a high confidence level the behavior of our experimental spectra obtained from laser-induced fluorescence (LIF) measurements, and then to allow us to extrapolate the fluorescence signal amplitude in other conditions than those used in these experiments.

The present interdisciplinary study combines electromagnetics and combustion to unveil an original and basic experiment displaying a spontaneous flame instability that is mitigated as the non-premixed sooting flame experiences a magnetic perturbation. This magnetic instability mitigation is reproduced by direct numerical simulations to be further elucidated by a flow stability analysis. A key role in the stabilization process is attributed to the momentum and thermochemistry coupling that the magnetic force, acting mainly on paramagnetic oxygen, contributes to sustain.

A Laser Extinction Method has been set up to provide two-dimensional soot volume fraction field time history at a tunable frequency up to 70 Hz inside an axis-symmetric diffusion flame experiencing slow unsteady phenomena preserving the symmetry. Early studies that used parasite-infected interleukin-4 (IL-4) reporter animals led us to identify basophils as the primary source of IL-4 and hence propose the hypothesis that basophils trigger the development of antigen-specific T helper type 2 (Th2) immune responses in vivo. These findings appeared to resolve a long-standing puzzle underlying Th2 immunity, that is, 'what is the source of the initial IL-4 necessary for CD4 T-cell differentiation into Th2 effector cells?'. However, results from extensive investigations of the contribution of basophils to Th2 immunity unveiled some controversial data that cast doubt on the initial hypothesis.

Basophils are recognized as immune modulators through their ability to produce IL-4, a key cytokine required for Th2 immunity. It has also recently been reported that basophils are transiently recruited into the draining lymph node (LN) after allergen immunization and that the recruited basophils promote the differentiation of naive CD4 T cells into Th2 effector cells. Using IL-3(-/-) and IL-3Rbeta(-/-) mice, we report in this study that the IL-3/IL-3R system is absolutely required to recruit circulating basophils into the draining LN following helminth infection.

Background: We evaluated whether contrast-enhanced multidetector computed tomography (CE-MDCT) might characterize myocardial infarct (MI) with patterns similar to those obtained by contrast-enhanced magnetic resonance (CE-MR) and studied the underlying mechanisms.

Methods And Results: In vivo infarct characterization by CE-MDCT was shown to be feasible between 4 and 20 minutes after contrast injection in 7 pigs with MI. Subsequently, in 16 patients with acute MI and 21 patients with chronic MI, contrast patterns by CE-MDCT were related to CE-MR.

Reference scattering curves for polarization and intensity produced by aggregates and agglomerates of ethylene and kerosene soot are obtained for scattering angles in the 10-170 degrees range. The polarization measurements were obtained with the Propriétés Optiques des Grains Astronomiques et Atmosphériques instrument for particles that levitate in microgravity during parabolic flights and on the ground by an air draught technique. Atrial natriuretic peptide (ANP) receptors were identified on both murine bone marrow-derived stromal cell lines A-3 and ALC and primary cultured cells using [125I]ANP binding assays and Northern blot analyses. The binding of [125I] ANP to the stromal cells was rapid, saturable, and of high affinity. The dissociation constants between ANP and its receptors on these cells showed no difference among cell types, while maximal binding capacity values were different among cell types.

Interleukin-3 (IL-3)-dependent mast cell lines, upon stimulation by calcium ionophores or by Fc epsilon RI cross-linking, express mRNA for, and secrete, a distinct pattern of cytokines, similar to those secreted by cloned mouse T cells of the TH2 type.
