Article Synopsis
  • Advances in cancer immunotherapy face challenges with patient resistance and relapses, prompting exploration of bispecific antibodies like NI-3201, designed to enhance T-cell activation against tumors.
  • NI-3201 works by blocking the PD-L1/PD-1 pathway and providing additional T-cell stimulation through CD28, showing promising in vitro and in vivo results for tumor regression and immune memory.
  • Preclinical safety assessments indicate good tolerability, and future studies aim to further investigate NI-3201's potential in improving outcomes for patients with PD-L1+ solid tumors.

Background: Upper limb impairment post-stroke often leads to a predominant use of the less affected arm and consequent learned disuse of the affected side, hindering upper limb outcome. Wearable sensors such as accelerometers, combined with smart reminders (i.e.

Dysregulation of the renin-angiotensin-aldosterone-system (RAAS) in sepsis is a complex and early phenomenon with a likely significant contribution to organ failure and patient outcomes. A better understanding of the pathophysiology and intricacies of the RAAS in septic shock has led to the use of exogenous angiotensin II as a new therapeutic agent. In this review, we report a multinational and multi-disciplinary expert panel discussion on the role and implications of RAAS modulation in sepsis and the use of exogenous angiotensin II.

Article Synopsis
  • About 62.3% of critically ill patients admitted for sepsis developed acute kidney injury (SA-AKI) during their hospital stay.
  • The study found that severe SA-AKI significantly increased the risk of in-hospital mortality among these patients.
  • Proper hydration and fluid resuscitation within the first hour of treatment were linked to lower mortality rates in patients with severe SA-AKI.

Aims: Sebaceous neoplasms constitute a group of adnexal tumours, including sebaceous adenoma, sebaceoma and sebaceous carcinoma. Although mismatch repair deficiency may be observed, the nature of the genetic alterations contributing to the development of most of these tumours is still unknown. In the present study, we describe the clinical, microscopic, and molecular features of eight sebaceomas with GRHL gene rearrangement.

Acute kidney injury (AKI) is a multifactorial syndrome with a high risk of short- and long-term complications as well as increased health care costs. The traditional biomarkers of AKI, serum creatinine and urine output, have important limitations. The discovery of new functional and damage/stress biomarkers has enabled a more precise delineation of the aetiology, pathophysiology, site, mechanisms, and severity of injury.

Article Synopsis
  • * Conducted across 40 hospitals in France, the trial involved 2,222 patients who were randomly assigned to either keep using RASIs until surgery or stop them 48 hours prior.
  • * The results showed no significant difference in all-cause mortality or major complications within 28 days after surgery between the two groups, suggesting that either strategy can be safely applied.

Article Synopsis
  • The study explores the relationship between acute kidney injury (AKI) and myocardial injury (MI) in patients with severe burns, identifying a condition called cardiorenal syndrome (CRS).
  • Researchers measured biomarkers galectin-3 (Gal3) and soluble CD146 (sCD146) daily for the first week after admission to evaluate their effectiveness in predicting CRS.
  • Results indicated that elevated levels of Gal3 at admission and during the week were significantly associated with CRS, showing good predictive performance, while sCD146 was less reliable in this context.

Purpose: Novel interventions for the prevention or treatment of acute kidney injury (AKI) are currently lacking. To facilitate the evaluation and adoption of new treatments, the use of the most appropriate design and endpoints for clinical trials in AKI is critical and yet there is little consensus regarding these issues. We aimed to develop recommendations on endpoints and trial design for studies of AKI prevention and treatment interventions based on existing data and expert consensus.

Article Synopsis
  • Recent advancements in mechatronics have led to the development of advanced arm prostheses, but challenges remain in designing effective control schemes, particularly for those with higher levels of impairment.
  • Current commercial options are limited in versatility and customization, pushing researchers to use specialized lab apparatus for testing new innovations.
  • The Smart Arm platform was created as a customizable, multi-articulated robotic arm prosthesis that supports research on control and sensory feedback, demonstrating notable success in practical applications like the Cybathlon event.

Background: Interlimb transfer of sequential motor learning (SML) refers to the positive influence of prior experiences in performing the same sequential movements using different effectors. Despite evidence from intermanual SML, and while most daily living activities involve interlimb cooperation and coordination between the four limbs, nothing is known about bilateral SML transfer between the upper and lower limbs.

Research Question: We examined the transfer of bilateral SML from the upper to the lower limbs and vice versa.

Background: In burn patients, skin barrier disruption and immune dysfunctions increase susceptibility to invasive fungal diseases (IFDs) like invasive candidiasis (IC) and invasive mold infections (IMI). We provide an in-depth analysis of IFD-related factors and outcomes in a 10-year cohort of severe burn patients.

Methods: This retrospective cohort study includes adult patients admitted to the burn intensive care unit (BICU) between April 2014 and May 2023 with total burn surface area (TBSA) ≥15%.

Article Synopsis
  • * Low-dose corticosteroids (≤400 mg hydrocortisone equivalent daily) have been shown to reduce mortality in patients with severe pulmonary infections, including COVID-19, community-acquired pneumonia, and Pneumocystis pneumonia.
  • * Clinical trials indicate that treatments like dexamethasone for COVID-19 and hydrocortisone for pneumonia can lead to lower mortality rates compared to placebo, highlighting the effectiveness of these corticosteroids in critical care settings.

Purpose: The development of acute kidney injury (AKI) after the acute respiratory distress syndrome (ARDS) reduces the chance of organ recovery and survival. The purpose of this study was to examine the AKI rate and attributable mortality in ARDS patients.

Methods: We performed an individual patient-data analysis including 10 multicenter randomized controlled trials conducted over 20 years.

Article Synopsis
  • Porocarcinoma is a malignant sweat gland tumor that can develop from benign poromas, and recent studies have identified specific genetic fusions like PAK1/2/3 in some cases.
  • In a study of 12 porocarcinoma patients, most were older males with tumors located on various parts of the body, and some patients developed distant metastases.
  • The research indicates that PAK1/2/3 fusions might drive cancer development in porocarcinomas that do not have YAP1 rearrangements, highlighting a potential target for treatment.
