Meta-analysis of the efficacy and safety of vamorolone in Duchenne muscular dystrophy.

Background: Duchenne muscular dystrophy (DMD) is a severe neuromuscular disorder, often leading to wheelchair dependence by age 13 with limited treatment options, largely relying on glucocorticosteroids. We assessed the efficacy and safety of vamorolone, a modified synthetic corticosteroid, for DMD.

Methods: We performed a systematic review and meta-analysis using seven databases including prospective studies comparing vamorolone with glucocorticosteroids or placebo in DMD patients.

Background: Almost 40% of individuals at ultra-high risk (UHR) for psychosis experience persistent attenuated psychotic symptoms (APS) yet it is unclear (1) whether they share overlapping clinical and functional outcomes compared to individuals who transition to psychosis, (2) when symptom and functioning trajectories begin to diverge between UHR individuals with different clinical outcomes, and (3) whether non-remission (persistent APS or transition) can be predicted using baseline and/or longitudinal data.

Study Design: Participants were drawn from 2 randomized clinical trials: Neurapro (n = 220; discovery sample) and STEP (n = 180; external validation sample). First, 12-24 month symptoms and functioning were compared between UHR individuals with persistent APS, sustained remission, or transition to psychosis.

Salinization is one of the main pedogenetic processes occurring on the semiarid zone of the Weddel sea, like in Seymour Island, where salt-affected soils are widespread. This study aimed to investigate whether topography controls the distribution of salt-affected soils on Seymour Island, particularly with reference to the Lopez de Bertodano Formation. Chemical and physical results from 45 soil profiles were used, where morphometric variables were extracted.

The concept of Targeted Protein Degradation (TPD) has been introduced as an attractive alternative to the development of classical inhibitors. TPD can extend the range of proteins that can be pharmacologically targeted beyond the classical targets for small molecule inhibitors, as a binding pocket is required but its occupancy does not need to lead to inhibition. Revealing a previously unknown function for intracellular C3d, we found that C3d engaged T cell responses against malignant PC in the bone marrow of mice that had developed multiple myeloma spontaneously.

: Patients with a HeartMate 3 (HM3) left ventricular assist device (LVAD) typically receive anticoagulation and antiplatelet therapy. The HM3 has shown a marked reduction in hemocompatibility-related adverse events (HRAEs) like stroke, bleeding, and pump thrombosis. This study evaluated whether aspirin (ASA) response influences HRAE incidence and if ASA sensitivity changes over time in HM3 recipients.

Polygenic risk scores and embryonic screening: considerations for regulation.

Polygenic risk scores (PRSs) have recently been used to inform reproductive decision-making in the context of embryonic screening. While this is yet to be widespread, it is contested and raises several challenges. This article provides an overview of some of the ethical considerations that arise with using PRSs for embryo screening and offers a series of regulatory considerations for jurisdictions that may wish to permit this in the future.

Musicality and social cognition in dementia: clinical and anatomical associations.

Human musicality might have co-evolved with social cognition abilities, but common neuroanatomical substrates remain largely unclear. In behavioural variant frontotemporal dementia, social cognitive abilities are profoundly impaired, whereas these are typically spared in Alzheimer's disease. If musicality indeed shares a neuroanatomical basis with social cognition, it could be hypothesized that clinical and neuroanatomical associations of musicality and social cognition should differ between these causes of dementia.

Streamlining the Highly Reproducible Fabrication of Fibrous Biomedical Specimens toward Standardization and High Throughput.

Soft nano- and microfiber-based polymer scaffolds bear enormous potential for their use in cell culture and tissue engineering since they mimic natural collagen structures and may thus serve as biomimetic adhesive substrates. They have, however, so far been restricted to small-scale production in research labs with high batch-to-batch variation. They are commonly produced via electrospinning or melt electrowriting and their delicate nature poses obstacles in detachment, storage, and transportation.

Increasing the utility of epidemiologic studies as key evidence in chemical risk assessment.

The Society of Toxicology 2024 meeting assembled risk assessors, epidemiologists, and toxicologists to discuss the utility of integrating epidemiologic data into the derivation of reference values. Advantages of the use of epidemiologic evidence include (i) human relevance; (ii) increased likelihood that exposure levels are relevant to risk assessment; and (iii) incorporation of uncertainties attributed to co-exposures or other population-based considerations. The workshop panelists discussed the challenges of incorporating epidemiologic evidence due to uncertain exposure measurements, confounding, heterogeneity, and inherent study design limitations.

Ring size effects on geometries and electronic structures were investigated for the (C H )M(C H ) ( = 4, 5, or 6; = 8, 7, or 6; + = 12; M = Ti-Ni) systems using density functional theory. The lowest-energy CHM structures for the early transition metals titanium, vanadium, and chromium are the experimentally known singlet (η-CH)Ti(η-CH), doublet (η-CH)V(η-CH), and singlet (η-CH)Cr, respectively. The likewise experimentally known singlet (η-CH)Ti, doublet (η-CH)V, and singlet (η-CH)Cr(η-CH) are the second-lowest-energy structures with only a small energy difference between the two vanadium structures.

A descriptive analysis of sarcopenia markers in young adults with down syndrome.

Background: Most body composition research in individuals with Down Syndrome (DS) has focused on overweight and obesity. Although limited research has shown muscle mass indexes and physical performance levels of adults with DS are similar to or lower than older adults with sarcopenia, there is still a large gap in the literature regarding sarcopenia.

Objective: This brief report provides a comprehensive descriptive analysis of sarcopenia and obesity markers in individuals with DS.

An injury-induced mesenchymal-epithelial cell niche coordinates regenerative responses in the lung.

Severe lung injury causes airway basal stem cells to migrate and outcompete alveolar stem cells, resulting in dysplastic repair. We found that this "stem cell collision" generates an injury-induced tissue niche containing keratin 5 epithelial cells and plastic Pdgfra mesenchymal cells. Single-cell analysis revealed that the injury-induced niche is governed by mesenchymal proliferation and Notch signaling, which suppressed Wnt/Fgf signaling in the injured niche.

Associations Between Oncology Outreach and Patient-Sharing Measures of Care Coordination.

Background: Oncology outreach is a common strategy for addressing cancer workforce shortages, where traveling oncologists commute across clinical settings to extend their services. Despite its known benefits specifically for rural patients, oncology outreach reallocates physician resources to satellite clinics and may negatively impact the coordination of cancer care.

Methods: In this retrospective study, we identified patients with incident breast, colorectal, and lung cancers from 2016-2019 nationwide Medicare claims and linked them to oncologists using Part B.

Vascularized lymph node transfer (VLNT) versus lymphaticovenous anastomosis (LVA) for chronic breast cancer-related lymphedema (BCRL): a retrospective cohort study of effectiveness over time.

Purpose: Microsurgical reconstruction, including vascularized lymph node transfer (VLNT) and lymphaticovenous anastomosis (LVA), have emerged as promising treatment options for chronic breast cancer-related lymphedema (BCRL). Despite their clinical relevance, the precise timelines for patient improvement following these interventions remain rather unexplored. Therefore, the goal of this study was to compare the long-term outcomes and improvement patterns over time of VLNT versus LVA to lay open potential differences and aid in personalized counseling of future patients.
