Article Synopsis
  • Mutations in the KRAS protein are common in cancer, with a notable example being the G12C mutation, which has targeted treatments.
  • Researchers developed a novel small molecule that can degrade many KRAS mutations, demonstrating more effective and lasting effects compared to traditional inhibition.
  • Their findings showed that this approach not only effectively kills cancer cells with KRAS mutations but also spares normal cells, leading to reduction in tumors in animal models.

Article Synopsis
  • A 70-year-old woman with metastatic non-small cell lung cancer (NSCLC) initially responded to erlotinib, but after progression, she was treated with platinum/pemetrexed and had a favorable outcome for over two years.
  • After further testing of her cancer revealed new mutations alongside an existing one, she transitioned to crizotinib, achieving a 17-month response before it was combined with afatinib, leading to another two-year improvement.
  • This case highlights the importance of ongoing molecular testing in metastatic NSCLC, as it can uncover new mutations that enable more effective treatment options and enhance patient survival.

Background: Head tremor is common in dystonia syndromes and difficult to treat. Deep brain stimulation (DBS) is a therapeutic option in medically-refractory cases. In most DBS-centers, the globus pallidus internus (GPi) is targeted in patients with predominant dystonia and the ventrointermediate nucleus of the thalamus (Vim) in predominant tremor.

Introduction: Thalamic gliomas pose a particular therapeutic challenge as complete resection is rarely achieved due to the deep and eloquent location. Laser interstitial thermal therapy (LITT) may provide a valuable management option for deep-seated gliomas that are not accessible with open surgery.

Case Presentation: A 57-year-old woman presented with a rapidly progressive large thalamic glioblastoma.

The open-face design of the Leksell Vantage frame provides many advantages. However, its more rigid, contoured design offers less flexibility than other frames. This is especially true for posterior fossa approaches.

Treatment with sotorasib has shown intracranial complete responses and continued intracranial stabilization in mutated non-small-cell lung carcinoma (NSCLC) patients with previously treated, stable brain metastases in a post hoc analysis of the ongoing CodeBreaK 100 trial. We present the case of a patient with mutant adenocarcinoma of the lung with active untreated brain metastases with a nearly complete intracranial response only 6 weeks after start of sotorasib illustrating the benefit of sotorasib in patients with active, previously untreated brain metastases in mutated NSCLC.

The class I histone deacetylases are essential regulators of cell fate decisions in health and disease. While pan- and class-specific HDAC inhibitors are available, these drugs do not allow a comprehensive understanding of individual HDAC function, or the therapeutic potential of isoform-specific targeting. To systematically compare the impact of individual catalytic functions of HDAC1, HDAC2 and HDAC3, we generated human HAP1 cell lines expressing catalytically inactive HDAC enzymes.

International data suggest that exposure to nature is beneficial for mental health and well-being. The restrictions related to the COVID-19 pandemic have created a setting that allows us to investigate the importance of greenness exposure on mental health during a period of increased isolation and worry. Based on 2060 responses from an online survey in Stockholm County, Sweden, we investigated: (1) whether the COVID-19 pandemic changed peoples' lifestyle and nature-related habits, and (2) if peoples' mental health differed depending on their exposure to greenness.

It is a physician's certified continuing medical education category-rather than their actual medical activity, in this case in the field of trauma surgery-that is decisive in filling statutory health insurance (SHI) practice vacancies. This evaluation arising from §16 of the requirement planning guideline applies accordingly when filling physician vacancies. Thus physician vacancies or statutory health insurance (SHI) practice places can only be filled by a physician in the same physician group in line with requirement planning.

Presentation of non-aversive light stimuli for several seconds was found to reliably induce locomotor activation and exploratory-like activity. Light-induced locomotor activity (LIA) can be considered a convenient simple model to study sensory-motor activation. LIA was previously shown to coincide with serotonergic and dopaminergic activation in specific cortical areas in freely moving and anesthetized animals.

A novel approach towards the asymmetric synthesis of N-stereogenic molecules via double aza-Michael addition was developed. The diastereomeric ratio can be increased by a thermodynamically controlled isomerization mechanism. Simple separation and functionalization of the products afford N-stereogenic compounds in high enantiomeric purity.

Primary emissions from a log wood burner and a pellet boiler were characterized by online measurements of the organic aerosol (OA) using a high-resolution time-of-flight aerosol mass spectrometer (HR-TOF-AMS) and of black carbon (BC). The OA and BC concentrations measured during the burning cycle of the log wood burner, batch wise fueled with wood logs, were highly variable and generally dominated by BC. The emissions of the pellet burner had, besides inorganic material, a high fraction of OA and a minor contribution of BC.

This study reports the potential toxicological impact of particles produced during biomass combustion by an automatic pellet boiler and a traditional logwood stove under various combustion conditions using a novel profluorescent nitroxide probe, BPEAnit. This probe is weakly fluorescent but yields strong fluorescence emission upon radical trapping or redox activity. Samples were collected by bubbling aerosol through an impinger containing BPEAnit solution, followed by fluorescence measurement.

Article Synopsis
  • Maternal deprivation (MD) is studied to understand how early adverse experiences affect behavior and neurobiology, potentially modeling major depression in animals.
  • In the study, male Long-Evans rats showed increased behavior transitions and sucrose preference due to MD, while female rats exhibited reduced swimming activity and cognitive performance.
  • The findings suggest MD does not strongly mimic major depression symptoms but may be more indicative of dysthymia, particularly in females, reflecting issues related to maternal relationships.

Neurokinin(3) receptors (NK(3)-Rs) have been implicated in psychomotor activity and reinforcement mechanisms. Recently, we showed that NK(3)-R antagonism blocked the psychostimulant properties of cocaine both in rats and in primates. Here, using in vivo microdialysis in the nucleus accumbens (NAc) of freely moving rats, we investigated the effect of the NK(3)-R agonist senktide (0.

The receptor-binding factor (RBF) for the avian oviduct progesterone (Pg) receptor (PR) has previously been shown to be a unique 10-kDa nuclear matrix protein that generates high affinity PR-binding sites on avian DNA. This paper describes the use of Southwestern blot and DNA gel shift analyses with RBF protein to identify a minimal 54-base pair RBF-binding element in the matrix-associated region (MAR) of the Pg-regulated c-myc gene promoter. This element contains a 5'-GC-rich domain and a 3'-AT-rich domain, the latter of which has a homopurine/homopyrimidine structure.
