Article Synopsis
  • - Preeclampsia is a pregnancy-related condition with unclear causes, and the protein irisin, linked to exercise, may play a role in its development and the health of the placenta.
  • - Research shows that pregnant individuals with preeclampsia have lower irisin levels, which appear early and correlate with other health indicators like blood pressure and protein levels.
  • - Increasing irisin levels in lab studies improved certain functions of trophoblast cells (which support embryo development), while lower levels led to symptoms similar to preeclampsia in animal models.

  • - Cabotegravir, used for HIV treatment and prevention via intramuscular injection, is being evaluated for thigh muscle as an alternative injection site compared to the traditional gluteal site, with a focus on its pharmacokinetics and demographic impacts.
  • - In a study involving both HIV-negative and HIV-positive participants, researchers collected and analyzed cabotegravir concentration data from thigh and gluteal injections, finding significant differences in absorption rates based on sex and body mass index (BMI).
  • - The analysis indicates that while thigh injections have a slower absorption rate and slightly lower bioavailability compared to gluteal injections, the results provide important insights for optimizing dosing strategies and future research on cabotegravir use.

Poliovirus (PV) is on the brink of eradication due to global vaccination programs utilizing live-attenuated oral and inactivated polio vaccines. Recombinant PV virus-like particles (VLPs) are emerging as a safe next-generation vaccine candidate for the impending polio-free era. In this study, we investigate the production, antigenicity, thermostability, immunogenicity, and structures of VLPs derived from PV serotype 2 (PV2) wildtype strain and thermally stabilized mutant (wtVLP and sVLP, respectively).

Unlabelled: HPTN 084 demonstrated the superiority of long-acting injectable cabotegravir (CAB-LA) compared with daily oral tenofovir disoproxil fumarate-emtricitabine (F/TDF) for HIV prevention in women. CAB-LA (600 mg) or placebo injections were administered 4 weeks after an initial dose (loading dose) and every 2 months (Q2M) thereafter; this is the approved regimen. Participants experienced both loading dose and Q2M delays during the trial.

Renal sympathetic nerves play a crucial role in the pathogenesis of hypertension, and renal denervation (RDN) is a new solution for patients with refractory hypertension. However, current RDN techniques show inconsistent results in clinical application probably owing to incomplete endovascular ablation of the sympathetic nerves and a lack of measures to localize and assess efficacy. In this study, a closed-loop RDN system consisting of a sensing unit with a piezoelectric thin-film sensor (PTFS) and a treatment unit with a hollow Pd nanoparticle shell (PdNPS) with a diameter of 202.

  • In June 2019, a patient in Inner Mongolia developed severe symptoms after a tick bite, leading to the discovery of a new virus named Wetland virus (WELV) through advanced genetic testing.
  • Active surveillance identified 17 cases of WELV infection across multiple regions in China, with patients exhibiting a range of nonspecific symptoms and laboratory abnormalities.
  • WELV was isolated from various ticks and animals, and studies showed that it can cause serious illness in mice, indicating a potential tick vector for the virus.

Unlabelled: Long-acting cabotegravir is approved for pre-exposure prophylaxis and combination HIV treatment, both initiated with optional short-term oral lead-in (OLI). We evaluated the impact of OLI on long-acting cabotegravir pharmacokinetics. Cabotegravir plasma concentrations were compared between HIV-positive participants initiating injections with ( = 278) or without ( = 110) OLI in phase III treatment study FLAIR and in HIV-negative participants using OLI ( = 263) in pivotal pre-exposure prophylaxis studies HPTN 083 and HPTN 084.

Background And Objectives: Exome sequencing (ES) demonstrates a 20-50 percent diagnostic yield for patients with a suspected monogenic neurologic disease. Despite the proven efficacy in achieving a diagnosis for such patients, multiple barriers for obtaining exome sequencing remain. This study set out to assess the efficacy of ES in patients with primary neurologic phenotypes who were appropriate candidates for testing but had been unable to pursue clinical testing.

Context: Clinical practice guidelines recommend palliative care for people with advanced heart failure (aHF), yet it remains underutilized.

Objectives: We examined medical center variation in specialist palliative care (SPC) and identified factors associated with variation among people with aHF.

Methods: We conducted a retrospective cohort study of 21,654 people with aHF who received healthcare in 83 Veterans Affairs Medical Centers (VAMCs) from 2018-2020.

Aims: A therapeutic strategy for chronic heart failure (HF) is to lower resting heart rate (HR). Ivabradine is a well-known HR-lowering agent, but limited prospective data exist regarding its use in Chinese patients. This study aimed to evaluate the effectiveness and safety of ivabradine in Chinese patients with chronic HF.

Background: The pathogenesis of chronic thromboembolic pulmonary hypertension (CTEPH) is multifactorial and growing evidence has indicated that hematological disorders are involved. Clonal hematopoiesis of indeterminate potential (CHIP) has recently been associated with an increased risk of both hematological malignancies and cardiovascular diseases. However, the prevalence and clinical relevance of CHIP in patients with CTEPH remains unclear.

Article Synopsis
  • Cabotegravir + rilpivirine is the first long-acting HIV treatment approved for intramuscular administration, typically in the gluteal area, but the lateral thigh is being explored as an alternative injection site.
  • A study assessed the safety and pharmacokinetics of this treatment in healthy adults after they received initial oral doses followed by thigh injections, with evaluations continuing for 52 weeks post-injection.
  • Results showed that thigh injections had similar tolerability and effectiveness compared to gluteal injections, with the most common side effects being mild pain and swelling at the injection site.

Background: Papillary thyroid cancer (PTC) is one of the most common endocrine malignancies with different risk levels. However, preoperative risk assessment of PTC is still a challenge in the worldwide. Here, the authors first report a Preoperative Risk Assessment Classifier for PTC (PRAC-PTC) by multidimensional features including clinical indicators, immune indices, genetic feature, and proteomics.

Glaucoma, a leading cause of irreversible blindness, is a highly heritable human disease. Previous genome-wide association studies have identified over 100 loci for the most common form, primary open-angle glaucoma. Two key glaucoma-associated traits also show high heritability: intraocular pressure and optic nerve head excavation damage quantified as the vertical cup-to-disc ratio.

The objective of this study is to describe the separation and identification of one new phenolic and 19 known compounds from . Their structures were determined based on spectroscopic (NMR, CD, and MS) data analysis or Mosher's method, and were compared with those reported in the literature. These isolates were then evaluated for their anti-inflammatory and antioxidant activities based on the inhibition of nitric oxide (NO) and interleukin (IL)-8 production in lipopolysaccharide (LPS)-stimulated cells (RAW264.
