Purification of bacterial endotoxins by zonal centrifugation.

In this paper, we describe a cultivation procedure in large-capacity fermentors (200 liters) and the purification of the Salmonella crude extracts by means of zonal centrifugation in a sucrose gradient. Briefly, the endotoxin has been extracted from salmonellae with hypertonic solutions. The crude endotoxin was shown to contain several antigens, mainly r(1), r(2), r(3), or heterologous antigen.

Isolation of Intact Chloroplasts from Euglena gracilis by Zonal Centrifugation.

Chloroplasts were separated from Euglena gracilis by zonal centrifugation at low speed in density gradients of Ficoll or dextran. The chloroplasts were intact by the criteria of ultrastructure and their content of ribulose diphosphate carboxylase and soluble protein. The chloroplasts also contained ribosomes and ribosomal RNA uncontaminated by the corresponding cytoplasmic particles.

Isolation of nuclei from a marine dinoflagellate.

By means of a medium containing dextran, nuclei were isolated in high yield from cells of Gymnodinium nelsoni, a marine dinoflagellate. Most of the DNA, but less than onetenth of the RNA, of the original cells was recovered in the purified nuclei. The nuclei appeared substantially intact as observed by light or electron microscopy.
