Small (SK) and intermediate (IK) conductance calcium-activated potassium channels are candidate ion channels for the regulation of excitability in nociceptive neurones. We have used unique peptide-directed antisera to describe the immunocytochemical distribution of the known isoforms of these ion channels in dorsal root ganglia (DRG) and spinal cord of the rat. These investigations sought to characterize further the phenotype and hence possible functions of nociceptive neurone subpopulations in the rat.

Immunocytochemistry exploits the incomparable specificity of the antibody-antigen interaction to form the basis of a flexible approach to the study of expression and localization of proteins both in model systems and their physiological context. This chapter details the theory and practice of the technique as well as lists the materials required. A general protocol is proposed, which can be adapted to suit the needs of individual investigators using the suggestions outlined in the Notes.

View Article and Find Full Text PDFA method for culturing primary rat alveolar macrophages (AMs) for 14 days was used to compare their responses to crocidolite and chrysotile asbestos fibres. Exposure to crocidolite increased production of tumour necrosis factor-alpha (TNF-alpha) and interleukin 1beta (IL-1beta), whereas exposure to chrysotile did not; neither fibre altered the production of interleukin 6 (IL-6). IL-1beta production increased progressively, while TNF-alpha was fully elevated from day 1.
