We have dissected the state of fibrinolytic balance in the C57/BL mouse macrophages, by means of immunotrap assays and zymography. We have monitored the individual changes of plasminogen activator (PA) and plasminogen activator inhibitor (PAI) activities of cellular lysates and secretions of these macrophages, after they were stimulated by various exogenous agents. The resident peritoneal macrophages were found to have very little PA but high level of PAI, and are therefore highly anti-fibrinolytic in nature.

The cDNA coding for a novel protein kinase from soybean (Glycine max L.), named GmPK6, was sequenced. The primary sequence of GmPK6 consists of 462 amino acids with an N-terminal sequence similar to the central region of Xenopus U1 snRNP 70K protein, and a C-terminal kinase domain representing structural mosaicism with features diagnostic of both protein serine/threonine and tyrosine kinases in eukaryotic organisms.

The RNA-dependent RNA polymerases (RdRp) of cucumber mosaic virus (CMV) and peanut stunt virus (PSV), members of the cucumovirus group, have been purified from virus infected plants and were used to study RNA synthesis in vitro using different viral RNAs, two cucumoviral satellites, and chimeric satellite cDNA clone transcripts as templates. The results show that solubilized RdRp preparations of CMV and PSV have a high degree of template dependency and catalyze (-) strand synthesis of the homologous cucumoviral RNAs with greater efficiency than the RNAs of heterologous cucumoviruses, although the PSV RdRp exhibits a lesser specificity than the CMV RdRp. On the other hand, both (-) and (+) strands of the satellite RNAs of CMV and PSV are synthesized by their homologous but not by the heterologous viral RdRps, indicating that recognition of satellites by the viral RdRp determines their replicative dependence upon specific helper viruses.

An insertion element, ISAE1, was discovered during the molecular analysis of mutants defective in the autotrophic growth (Aut-) of Alcaligenes eutrophus H1-4, a mitomycin C-generated derivative of strain H1. ISAE1 is 1,313 bp long, has 12-bp nearly perfect inverted terminal repeats, and contains an open reading frame that has a coding capacity of 408 amino acids. Direct repeats of 8 bp were generated by insertion of ISAE1 into chromosomes or plasmids.

A hierarchical system for character recognition with hidden Markov model knowledge sources which solve both the context sensitivity problem and the character instantiation problem is presented. The system achieves 97-99% accuracy using a two-level architecture and has been implemented using a systolic array, thus permitting real-time (1 ms per character) multifont and multisize printed character recognition as well as handwriting recognition.

Glycosylation-site-containing peptides were isolated from the proteolytic digests of luffin-a, luffin-b, PAP-S and CNBr-fragments of PAP-S by reverse-phase HPLC, and their amino acid compositions and sequences were analyzed. Six peptides were obtained from luffin-a, and three each from luffin-b and PAP-S. All of these peptides were negative toward the phenol-H2SO4 reaction and gave only N-acetyl-D-glucosamine in gas chromatography after methanolysis and reacetylation.

The complete amino acid sequence of antiviral protein from the seeds of pokeweed (Phytolacca americana) has been determined. This has been done by the sequence analysis of peptides derived by enzymatic digestion with trypsin, pepsin, and lysylendopeptidase, as well as by chemical cleavage with cyanogen bromide. The protein consists of 261 amino acid residues containing two disulfide bonds and has a calculated molecular mass of 29167 Da.
