Natural killer (NK) cells from patients with myeloid malignancies display global dysfunction with impaired killing capacity, altered metabolism, and an exhausted phenotype at the single-cell transcriptomic and proteomic levels. TCR repertoires expand and primarily emerge from CD8 cells in patients responding to treatment or having a stable disease, while TCR repertoires contract in therapyresistant patients.

Here, we show that PRC2 loss confers a dedifferentiated early neural-crest phenotype which is exclusive to PRC2-mutant MPNSTs and not a feature of neurofibromas.
