Colorectal cancer is a leading cancer in many countries. The glucose metabolic activities of the tumor cells are higher than that of the normal cells. They could uptake much more 2-[18F] Fluoro-2-Deoxy-d-Glucose (18F-FDG), an analogue of glucose, than normal cells do.

Pollen and seeds share a developmental sequence characterized by intense metabolic activity during reserve deposition before drying to a cryptobiotic form. Neither pollen nor seed development has been well studied in the absence of gravity, despite the importance of these structures in supporting future long-duration manned habitation away from Earth. Using immature seeds (3-15 d postpollination) of Brassica rapa L.

TRAIL receptor (TRAIL-R) signaling has been implicated in inducing apoptosis in tumor cells, but little is understood about its physiological function. Here, we report the generation and characterization of TRAIL-R(-/-) mice, which develop normal lymphocyte populations but possess enhanced innate immune responses. TRAIL-R(-/-) mice exhibited increased clearance of murine cytomegalovirus that correlated with increased levels of IL-12, IFN-alpha, and IFN-gamma.

A 15-mer phosphorothioate antisense oligonuclide (ASON) complementary to the translation start region of the C-myc oncogene mRNA was radioiodinated to enhance its antitumor activity, and vasoactive intestinal peptide bound covalently polylysine (VIP-polylysine) was used as a carrier to deliver the oligonucleotide into VIP receptor-positive tumor cells. The antitumor activity of radioiodinated ASON conjugated to VIP-polylysine(VIP-131I-ASON) was investigated in athymic mice bearing HT29 tumor xenografts in comparison with unconjugated radioiodinated ASON(131I-ASON), unlabelled ASON (VIP-ASON) and scrambled oligonucleotide (VIP-131I-MON) conjugated to VIP-polylysine. Conjugation 125I-ASON to VIP-polylysine resulted in a 5.

Background: Although a number of studies have discussed rhinoplasty of the Asian nose, few have focused specifically on the Asian-American nose, which incorporates characteristics from various Asian nationalities and/or ethnic groups.

Objective: The authors propose a classification system for aesthetic correction of the Asian-American nose and outline surgical techniques with general application to this patient population.

Methods: Patients were classified into 3 categories based on morphologic characteristics, as follows: type I (vertical deficiency), type II (vertical deficiency and broad nasal width), and type III (type I or II with glabella deficiency).

Objective: Based on the rationale that the ablation of thyroid remnant can effectively reduce the risk for recurrence of differentiated thyroid carcinoma (DTC) and hence decrease the case fatality rate. This randomized controlled trial was designed to assess the value of hydrochlorothiazide in the ablation of thyroid remnant with 131I.

Methods: Thirty consecutive DTC patients with thyroid remnant after thyroid surgery were divided into two groups by randomization, the hydrochlorothiazide group received hydrochlorothiazide 25 mg tid for 4 days, the control group received placebo.

Aim: To characterize the receptor binding affinity and cytotoxicity of insulin-methotrexate (MTX) for the potential utilization of insulin as carriers for carcinoma target drugs.

Methods: MTX was covalently linked to insulin. Insulin-MTX conjugate was purified by Sephadex G-25 column and analyzed by high performance liquid chromatography.

Since the introduction of percutaneous transluminal coronary angioplasty, restenosis has remained the most challenging problem facing interventional cardiologist. Intravascular radiation is a feasible and promising adjunctive therapy in restenosis treatment by suppressing both neointimal proliferation and constrictive remodeling, while there are growing concerns about its long-term effects and complications in clinical perspectives as well as dosing and paradoxical stimulation. Current comments on them may well favor the choice of comprehensive treatment protocol for clinicians.

DNA repair via the homologous recombination pathway requires the recombinase RAD51 and, in vertabrates, five RAD51 paralogs. The paralogs form two complexes in solution, a XRCC3/RAD51C heterodimer and a RAD51B/RAD51C/RAD51D/XRCC2 heterotetramer. Mutation of any one of the five paralog genes prevents subnuclear assembly of recombinase at damaged sites and renders cells 30-100 fold sensitive to DNA cross-linking drugs.

Objective: To explore the radiolabeling property of oligonucleotide with 99mTc using NHS-MAG3 as a bifunctional chelator.

Methods: Three 15-base single-stranded amine-derivitized oligonucleotides, which were antisense(ASON), sense(SON) and mismatched oligonucleotides(MON) of c-myc oncogene mRNA, were coupled with NHS-MAG3 and labeled with 99mTc. The labeled oligonucleotide was purified by Sephadex G25 column chromatogram, then the stability was evaluated.

TSH secretion is controlled by the stimulatory action of hypothalamic TRH and the inhibition via central dopaminergic and somatostatinergic mechanisms as well as by a hypothalamic inhibitory action of thyroid hormones. Metoclopramide is DA2 receptor blockade which can accelerate to elevate the concentration of serum TSH in hypothyroidism, which is accompanied by a progressive loss of dopaminergic tone as demonstrated by a progressive increase in prolaction serum levels, especially in patients with differentiated thyroid carcinoma(DTC). So the period of L-thyroxine withdrawal is contracted and the symptoms of hypothyroidism is released.

The results of study of Brassica embryo differentiation and reserve nutrient substance accumulation in the seeds were represented. Near resemblance of the spaceflight and around control embryo development was revealed. Different character of the reserve substance accumulation was noted, despite of the morphologic similarity in seeds produced in spaceflight and on the ground.

Reproductive development in microgravity has now been studied in a variety of plants; Arabidopsis, Brassica, and Triticum have been especially well studied. Earlier indications that gravity might be required for some stage of reproductive development have now been refuted. Nevertheless, the spaceflight environment presents many unique challenges that have often compromised the ability of plants to reproduce.

Aim: To evaluate the possibility of using insulin as a carrier for carcinoma-targeted therapy mediated by receptor, and to investigate the expression of insulin receptor in human hepatocellular carcinoma and the receptor binding characteristics of insulin-IUdR (iododeoxyuridine).

Methods: IUdR was covalently conjugated to insulin. Receptor binding assays of (125)I-insulin to human hepatocellular carcinoma and its adjacent tissue were performed.

Differentiated thyroid cancer can be effectively treated with high-dose 131I and the other head and neck cancer can also be effectively treated with extra-radiotherapy, but these treatments often result in a reduction in salivary gland function, causing xerostomia. Collectively, these effects can lead to severe secondary complications, including difficulty in speaking and swallowing, decreasing appetite even affecting nutrition and sleep. Amifostine, an analog of cysteamine, is a phosphorlyated aminothiol prodrug and its active metabolite, WR-1065 etc, can selectively protect normal tissues from the cytotoxic effects of drugs and/or radiation while preserve antitumor effects.

[Prospects for antisense techniques].

Sheng Wu Yi Xue Gong Cheng Xue Za Zhi

December 1999

Antisense technique, developed on the principle of base complementation, is a method using short strand oligonucleotide fragments complementary to target DNAs or target RANs for inhibition of gene expression. As a theoretically perfect and specific method to block the expression of gene, it has been rapidly developed over the last twenty years. This review expatiates on its techniques, its problem and possible way of problem-solving, and its recent application in medicine.
