Tumor-stroma ratio (TSR) has been recognized as a valuable prognostic indicator in various solid tumors. This study aimed to examine the clinicopathologic relevance of TSR in Merkel cell carcinoma (MCC) using artificial intelligence (AI)-based parameterization of the stromal landscape and validate TSR scores generated by our AI model against those assessed by humans. One hundred twelve MCC cases with whole-slide images were collected from 4 different institutions.

Article Synopsis
  • - Claudin-4 is crucial for tight junctions in the skin, contributing to the epidermal barrier and regulating movement between cells.
  • - The study analyzed 35 blistering disorders and found varying levels of claudin-4 expression, particularly in conditions like pemphigus vulgaris and bullous pemphigoid.
  • - Results indicate that claudin-4 may increase as a response to barrier dysfunction in these disorders, potentially revealing underlying mechanisms affecting skin integrity.

Programmed cell death protein 1 (PD-1) plays a pivotal role in immune system regulation, with its expression levels linked to malignancy prognosis. However, existing reports on PD-1 staining in mycosis fungoides (MF) present conflicting findings, and little attention has been given to PD-1 staining in different MF variants. To address this, we conducted a retrospective study, employing immunohistochemistry to examine PD-1 expression in cases of folliculotropic MF and non-folliculotropic MF.

Article Synopsis
  • The study focuses on the use of the TRPS1 immunohistochemical stain as a marker for breast carcinomas and evaluates its expression in skin tumors with follicular characteristics, such as trichoblastoma, trichoepithelioma, and basal cell carcinoma.
  • Researchers conducted IHC studies on 13 trichoblastomas, 15 trichoepitheliomas, and 15 basal cell carcinomas, revealing that trichoblastomas and trichoepitheliomas exhibited higher TRPS1 positivity compared to basal cell carcinomas.
  • Additionally, TRPS1 staining patterns showed unique mesenchymal cell behaviors in trichoblastomas and trichoepitheliomas, suggesting that TRPS1 could serve

We illustrate a rare case of coexisting extramedullary manifestation of CMML and new onset follicular lymphoma within the same core-needle biopsy of a lymph node. We discuss the differences between extramedullary hematopoiesis and extramedullary manifestation of myeloid neoplasms. We also highlight the importance of generous tissue sampling and thorough examination of nodal tissue in the setting of an established myeloid neoplasm to avoid missing rare but possible nodal involvement.
