Aquatic pathogens often cannot tolerate drying, and thus their spread, and diversity across a landscape may depend on interactions between hydrological conditions and the movement of infected hosts. The aquatic fungus Batrachochytrium dendrobatidis (Bd) is a nearly ubiquitous pathogen of amphibians and particular lineages have been associated with host declines. By coupling amphibian surveys with molecular pathogen detection and genotyping techniques, we characterized the spatial dynamics and genetic diversity of Bd on a landscape containing both permanent and ephemeral ponds.

View Article and Find Full Text PDFPolymerase chain reaction (PCR) based on single primers of arbitrary nucleotide sequence provides a powerful marker system for genome analysis because each primer amplifies multiple products, and cloning, sequencing, and hybridization are not required. We have evaluated this typing system for the mouse by identifying optimal PCR conditions; characterizing effects of GC content, primer length, and multiplexed primers; demonstrating considerable variation among a panel of inbred strains; and establishing linkage for several products. Mg2+, primer, template, and annealing conditions were identified that optimized the number and resolution of amplified products.
View Article and Find Full Text PDFAn interspecific backcross between lab mice and Mus spretus was used to construct a multilocus map of Chromosome 17 consisting of 12 new anonymous loci and 9 anchor loci. In addition, 7 anonymous DNA loci were added to the Chr 17 map for the BXD strains. Although we were able to identify readily the most likely gene order in the interspecific backcross, we found no evidence for an unambiguous gene order using the BXD recombinant inbred strains.
