[Antitumor activity of L-asparaginase from Erwinia carotovora from against different leukemic and solid tumours cell lines].

We have studied dose- and time-dependent antitumor and cytotoxic effects of Erwinia carotovora L-asparaginase (ECAR LANS) and Escherichia coli L-asparaginase (MEDAC) on human leukemic cells and human and animal solid tumor cells. We determined the sensitivity of tumor cells to L-asparaginases, as well the effect L-asparaginases on cell growth rate, protein and DNA synthesis per se and with addition of different cytostatics. The data obtained demonstrated that ECAR LANS L-asparaginase suppressed growth of all tested solid tumor cells.

[Relationship between the expression levels of PAPP-A metalloproteinase and growth and transcriptional factors in endometrial cancer].

We have examined for the first time the relationship between the expression of PAPP-A metalloproteinase and insulin-like growth factors (IGF-I, IGF-II, VEGF) and transcription factors (NF-kappaB, HIF-1) playing an important role in pathogenesis of cancer. We also demonstrated a positive association between the level of PAPP-A metalloproteinase and the level of growth (VEGF and IGF-I) and transcription factors (NF-kappaB p50, NF-kappaB p65, HIF-1alpha). The current findings suggest an important role of PAPP-A in regulation of bioavailability of IGF-I, VEGF, activated forms of NF-kappaB, and alpha-subunits of HIF-1 in endometrial tumors.

Multiplication and isolation of hepatitis C virus in the NGUK-1 rat neurinoma neural cells.

Hepatitis C virus (HCV) strains were isolated from human sera in vitro. NGUK-1 rat neurinoma neural cells [1] were selected as the experimental model of HCV. The cells were infected after attaining the confluence.

[Antitumor activity of L-asparaginase from Yersinia pseudotuberculosis].

The cytotoxic activity of L-asparaginases from Yersinia pseudotuberculosis and from Erwinia carotovora were investigated in vitro using several tumor cells lines: Jurkat and Molt-4 (human T-lymphoblastic leukemia), MCF-7 (human breast adenocarcinoma), LnCap (human prostate carcinoma), NGUK1 (rat Gasser node neurinoma). E. coli L-asparaginase produced by "Medak" (Germany) was used as a reference.

Galactose oxidase immobilized on silica in an analytical determination of galactose-containing carbohydrates.

Galactose oxidase from Fusarium graminearum IMV-1060 adsorbed on, and covalently bound to, silica carriers has been used for analytical determinations of D-galactose and galactose-containing sugars. Using a flowing oxygen electrode of the Clark-type, sensor system for enzymatic analysis of water solutions of galactose-containing carbohydrates was made. Measurements were taken both in the pulse and continuous modes of a substrate flowing through a column with an immobilized biocatalyst.

The review deals with the basic characteristics of prion diseases of man and animals. It details the studies of some little known aspects of the pathogenesis of prion diseases and their lifetime diagnosis, conducted at the M. P.

[In vivo diagnosis of Creutzfeldt-Jakob disease].

The paper presents the data concerning usage of some original method of vital laboratory diagnostics of Creutzfeldt-Jacob disease that belongs to the group of prionic diseases. The method consisted in the inoculation of inoculative culture of rat Gasser ganglion's neurinoma by biologic materials investigated (serum and clot of blood) with the following passivation and investigation of the contaminated culture by means of both morphologic and electron microscopic methods. As an example of vital verificated case the wide pathomorphologic analysis of the biopsy sample of brain was presented.

Modulation of glutamate neurotoxicity in the transformed cell culture by monoamine oxidase inhibitors, clorgyline and deprenyl.

Addition of 30mM glutamate to the culture medium decreased growth of rat glioma C6 cells accompanied by a decrease of DNA synthesis and an increase of lactate dehydrogenase (LDH) detected in the conditioned medium. The presence of 1 microM deprenyl attenuated the glutamate effect on cell growth only during the first 24-48 h incubation and had a minor influence on the glutamate-induced decrease of DNA synthesis. Clorgyline (1 microM) potentiated glutamate-induced DNA synthesis during the first 24 h incubation without significant influence on the cell growth.

[Lifetime diagnosis of prion transmissive encephalopathies].

An original method for life-time diagnosis of Greutzfeldt-Jakob's disease, a neurodegenerative disease belonging to transmissive spongiform encephalopathies, is proposed. It consists in inoculation of the examined biological materials (blood serum and clot) into a continuous culture of the rat Gasser's node neurinoma, followed by passages and study of the inoculated culture by morphological methods and electron microscopy.

[New transplantable brain gliomas in rats].

The paper provides the characteristics of 11 original transplanted rat brain gliomas that are part of the collection of experimental tumors of the nervous system, which has been developed at the Institute of Human Morphology, Russian Academy of Medical Sciences. The biological features of brain tumour strains, methods of their transplantation and cryoconservation are outlined in the paper.

[The Gerstmann-Sträussler syndrome: new diagnostic possibilities].

On the basis of clinical picture of the disease, data of CT, MRT, psychological study, original laboratory investigation aimed at indication of changes in transposed neuroglia cells induced by the causal agent of subacute spongious transmissible encephalopathies, the diagnosis of patient K., 49 years old, was considered to be: syndrome of Gertsmann-Sträussler [correction of Herstmann Streussler]. Duration of the disease was 2 years.

[The regional characteristics of the blood plasma lipid levels and of the indices of arterial pressure and physical development in schoolchildren of Russian nationality living in different climatic-geographic areas (based on the data from a single study in Ashgabad, Moscow, Orenburg, Tyumen and Tallinn)].

Random samples from a schoolchildren population living in different climatic and geographic regions were entered in the standard epidemiological survey. They were examined for plasma lipids vs. arterial pressure and physical development.

[Data for the substantiation of the maximum permissible exposure level of armos fiber dust in the air of the work area].

The 'Armos' fibre dusts are characterized by low cytotoxicity, absence of fibrogenicity, display non-toxic properties in single exposures with no skin-irritating and sensibilizing effects. In repeated introduction into the stomach, the trachea, and if inhaled, these dusts displayed weak toxic properties. Dust concentrations at 10 mg/m3 are in proximity to the threshold one.

[A new cell line of rat gasserian ganglion neurinoma NGUK-1].

A new cell line has been derived from the rat gasserian ganglion neurinoma NGUK-1 induced by transplacental administration of ethylnitrosourea. It is characterized by an astrocyte-like growth pattern at the low cell density, and by an epitheliocyte-like pattern in the confluent monolayer. The cell line displays a high proliferative activity, its maximum mitotic index and proliferative pool being--2.
