We developed a single-molecule enzyme activity assay platform for NAD(P)-dependent oxidoreductases, leveraging a new NAD(P)H-responsive fluorogenic probe optimized for microdevice-based fluorometric detection. This platform enabled the detection of enzyme activities in blood and cerebrospinal fluid (CSF), including lactate dehydrogenase, glucose-6-phosphate dehydrogenase, and hexokinases. We demonstrate its potential for activity-based diagnosis by detecting altered populations of enzyme activity species in blood and CSF from liver damage in brain tumor patients.

In surgery for acute type A aortic dissection, controlling bleeding from the posterior wall of the proximal anastomosis is particularly challenging. To address this, we use the "reversed turn-up technique." For the reinforcement of the proximal aortic stump, Teflon felt strips were placed inside and outside the suture line with 4-0 polypropylene continuous transverse mattress sutures, and BioGlue was applied to the false lumen.

Rupture of abdominal aortic aneurysm (AAA) due to an isolated type II endoleak (TIIEL) is rarely reported, accounting for less than 1% of all TIIELs; typically, rupture associated with TIIEL is accompanied by type I or type III endoleaks. We report a case of ruptured AAA secondary to TIIEL without any other types of endoleaks, occurring late after endovascular abdominal aortic repair (EVAR). A 77-year-old man with a history of EVAR 11 years earlier presented with abdominal pain.

Connecting two small molecules, such as ligands, fluorophores, or lipids, together via a linker with amide bonds is a widely used strategy to generate synthetic bifunctional molecules for various biological and biomedical applications. Such bifunctional molecules have been used in live-cell experiments under the assumption that they should be stable in cells. However, we recently found that a membrane-targeting bifunctional molecule, composed of a lipopeptide and the small-molecule ligand trimethoprim, referred to as mgcTMP, underwent amide-bond cleavage in mammalian cells.

Article Synopsis
  • - The study assessed how colonic diverticular bleeding (CDB) was managed and its clinical outcomes during the COVID-19 pandemic, comparing data from two time periods: before the pandemic (April 2017-March 2020) and during it (April 2020-March 2023).
  • - A total of 388 hospitalized patients were analyzed using propensity score matching, revealing that while the overall number of colonoscopies remained similar, there was a notable decrease in early colonoscopies and those done outside regular hours during the pandemic.
  • - Despite the reduction in early and after-hours colonoscopies, the risks of rebleeding or poor outcomes did not significantly increase during the pandemic, suggesting that patient outcomes

Objectives: It is recommended to adjust the dose of vancomycin (VCM) with a target area under the concentration-time curve (AUC) of 400-600 μg·h/mL. Factors that affect the deviation between AUCs are estimated from the trough value alone and the trough and peak values using practical AUC-guided therapeutic drug monitoring (PAT) for vancomycin. In this study, factors that affect AUC were evaluated.

Methaemoglobin (metHb) possesses inherent characteristics that facilitate reversible binding to hydrogen sulphide. Exogenous hydrogen sulphide supplementation imparts beneficial bioactive effects, including antioxidant and anti-inflammatory; hence, we hypothesized that the metHb-hydrogen sulphide complex could act as a hydrogen sulphide donor for medication. In this study, we prepared a hydrosulphide-metHb-albumin (HS-metHb-albumin) cluster and examined its applicability as a hydrogen sulphide donor in the mice model of hepatic ischemia-reperfusion injury.

Article Synopsis
  • The study investigates how ordinary people assess robots versus humans that make moral decisions, particularly in norm conflict scenarios like the trolley dilemma.
  • It involves 13 studies with 7,670 participants and explores a variety of factors that might influence evaluations, such as the type of moral judgment, cultural context, and aspects of empathy.
  • Findings suggest that while general moral norms are similar for both, humans face less blame than robots when it comes to inaction in critical decisions, possibly because people empathize with the difficult choices humans must make, a sentiment not extended to robots.
