Decreased activity and expression of the G-protein coupled receptor GPR88 is linked to many behavior-linked neurological disorders. Published preclinical GPR88 allosteric agonists all have pharmacokinetic properties that preclude their progression to the clinic, including high lipophilicity and poor brain penetration. Here, we describe our attempts to improve GPR88 agonists' drug-like properties and our analysis of the trade-offs required to successfully target GPR88's allosteric pocket.

Ferromagnetic ferrofluids are synthetic materials consisting of magnetic nanoplatelets dispersed in an isotropic fluid. Their main characteristics are the formation of stable magnetic domains and the presence of macroscopic magnetization even in the absence of a magnetic field. Here, the authors report on the experimental observation of spontaneous stripe formation in a ferromagnetic ferrofluid in the presence of an oscillating external magnetic field.

The reactions of aromatic aldehydes and levoglucosenone promoted by methoxide gives bridged α,β-unsaturated ketones, formed by a series of oxa-Michael-initiated cascade reactions in yields of up to 91% (14 examples). A complex series of equilibria operate during the reaction, and the formation of the bridged species is thermodynamically favored, except in the case of 5-methylfurfural and pyrrole-2-carboxaldehyde. This is the first report detailing this type of aldol/Michael cascade involving oxa-Michael initiation.

Article Synopsis
  • Researchers have developed a method to retrieve the refractive index profile of photonic structures using standard diffraction experiments and rigorous coupled-wave analysis, eliminating the need for additional data collection.
  • The technique involves selecting a probe wavelength for the diffraction that is significantly smaller than the structure's lattice constant, allowing full recovery of the phases of the Fourier components.
  • The study successfully measured the slight asymmetry of nanocomposite phase grating structures through both light and neutron diffraction methods.

Australia's endemic desert shrubs are commonly aromatic, with chemically diverse terpenes and phenylpropanoids in their headspace profiles. Species from the genus (Scrophulariaceae ex. Myoporaceae) are the most common, with 215 recognised taxa and many more that have not yet been described, widely spread across the arid parts of the Australian continent.

This paper reports imaging of objects with slow neutrons, specifically very cold neutrons and cold neutrons, at Institut Laue Langevin, using novel, permanent magnet (NdFeB) compound refractive lenses (MCRL) with a large 2.5 cm bore diameter. The MCRL focuses and images spin-up neutrons and defocuses spin-down neutrons via a large, radial magnetic field gradient.

We investigate the applicability of polymer-ionic liquid composites as optical elements for light, as well as for slow neutrons. The gratings are recorded using two-beam mixing and are characterized experimentally based on their diffraction properties. We produced a set of samples differing in their thickness, ranging from 10 m - 100 m .

A step-economic biomimetic synthesis of mitchellenes B-H found in has been achieved. Starting from the putative muurolane biological precursor, redox isomerization of the allylic alcohol gave an epimeric mixture of aldehydes, which could be used as a handle for cyclization onto the C6 position, using BuSnH-mediated radical cyclization or NHC-catalyzed Stetter reaction. The NHC-mediated approach was superior as the epimeric mixture underwent a dynamic kinetic resolution during the reaction, and reduction of the mixture with NaBH selectively formed the mitchellene ring system in 56% yield for the three steps.

Suspensions of magnetic nanoplatelets in isotropic solvents are very interesting examples of ferrofluids. It has been shown that above a certain concentration ΦNI such suspensions form a ferromagnetic nematic phase, which makes this system a unique example of a dipolar fluid. The formation of a nematic phase is driven by anisotropic electrostatic and long-range dipolar magnetic interactions.

Asymmetric 1,2-additions of cyanide yield enantioenriched cyanohydrins as versatile chiral building blocks. Next to HCN, volatile organic cyanide sources are usually used. Among them, cyanoformates are more attractive on technical scale than TMSCN for cost reasons, but catalytic productivity is usually lower.

Background: A multitude of troponin assays for the point-of-care (POC) have been developed showing a lack of analytical sensitivity and precision. We present a new platform solution for the high-sensitivity detection of cardiac troponin T (cTnT) in a 30 μL whole blood sample with a turnaround time of 11 min.

Methods: The immunoassay was completely run in a ready-to-use plastic disposable, a centrifugal microfluidic disc with fully integrated reagents.

Al-F bonds are among the most stable σ bonds known, exhibiting an even higher bond energy than Si-F bonds. Despite a stability advantage and a potentially high Lewis acidity of Al-F complexes, they have not been described as structurally defined catalysts for enantioselective reactions. We show that Al-F salen complexes with appended ammonium moieties give exceptional catalytic activity in asymmetric carboxycyanations.

We present an overview of recent investigations of photopolymerizable nanocomposite photonic materials in which, thanks to their high degree of material selectivity, recorded volume gratings possess high refractive index modulation amplitude and high mechanical/thermal stability at the same time, providing versatile applications in light and neutron optics. We discuss the mechanism of grating formation in holographically exposed nanocomposite materials, based on a model of the photopolymerization-driven mutual diffusion of monomer and nanoparticles. Experimental inspection of the recorded grating's morphology by various physicochemical and optical methods is described.

The strongly aromatic Australian desert species Eremophila dalyana is an Aboriginal medicinal plant that continues to be used today in Central Australia in the treatment of respiratory complaints and Sarcoptes scabiei infestation. Using hydrodistillation of aerial parts of the plant, the new natural product myodesert-l- ene was isolated in two disjunct populations at up to 98% of the volatiles present in the hydrodistilled oils. Weak antimicrobial activities were observed for whole oils and myodesert-l-ene.

We report influences of varying functionalities of thiols as chain transfer agents on the spatial frequency response, polymerization shrinkage, and thermal stability of a volume grating recorded in a photopolymerizable ZrO₂ nanoparticle-polymer composite film. It is shown that a substantial increase in the saturated refractive index modulation is realized at high spatial frequencies by doping with multifunctional thiols. Moreover, the incorporation of multifunctional thiols considerably suppresses polymerization shrinkage of recorded volume gratings and thermal changes in refractive index and film thickness as compared with the case of mono-thiol.

We demonstrate twofold enhancement of the saturated refractive index modulation (Δn(sat)) recorded in a photopolymerizable nanoparticle-acrylate polymer composite film by incorporating thiols acting as chain transfer agents. The chain transfer reaction of thiols with (meth)acrylate monomer reduces the polymer crosslinking density and facilitates the mutual diffusion of nanoparticles and monomer during holographic exposure. These modifications provide increased density modulations of nanoparticles and the formed polymer, resulting in the enhancement of Δn(sat) as high as 1.

The reference-geometry Harris-Foulkes (RGHF) approach has been used to model high-order terms within the expansion of multi-dimensional potential energy surfaces (PES) as needed within the calculation of accurate vibrational frequencies beyond the harmonic approximation. The key step of this method is a localization of the electron density to the atoms of the molecule at a given reference structure and a subsequent transfer of these atom-centered partial densities to the atom positions of distorted structures. This concept has been used to evaluate the 3-mode coupling terms of a multi-mode expansion of the PES as arising in the Watson Hamiltonian.

The geometric phase has been proposed as a candidate for noise resilient coherent manipulation of fragile quantum systems. Since it is determined only by the path of the quantum state, the presence of noise fluctuations affects the geometric phase in a different way than the dynamical phase. We have experimentally tested the robustness of Berry's geometric phase for spin-1/2 particles in a cyclically varying magnetic field.

In a neutron polarimetry experiment the mixed-state relative phases between spin eigenstates are determined from the maxima and minima of measured intensity oscillations. We consider evolutions leading to purely geometric, purely dynamical, and combined phases. It is experimentally demonstrated that the sum of the individually determined geometric and dynamical phases is not equal to the associated total phase which is obtained from a single measurement, unless the system is in a pure state.
