Expression of a CD44 variant and VEGF-C and the implications for lymphatic metastasis and long-term prognosis of human breast cancer.

The aim of the present study was to determine whether expression of the CD44 variant v7-v8 (CD44v7-v8) or vascular endothelial growth factor-C (VEGF-C) is associated with long-term prognosis in breast cancer patients. A 10-year follow-up of 91 patients with primary breast cancer who were previously assessed for CD44 expression was undertaken. Immunohistochemical evaluation of VEGF-C expression was performed in 87 of these patients and their long-term prognosis was assessed.

Efficacy of root canal preparation by Er,Cr:YSGG laser irradiation with crown-down technique in vitro.

Objective: The purpose of this study was to compare the efficacy of Er,Cr:YSGG laser in root canal preparation and its effectiveness in removing debris and smear layer with the efficacy of the conventional hand instrument, in vitro.

Background Data: Recently, the use of the Er,Cr:YSGG laser device in root canal preparation has been excepted in the dental clinic. However, there have been no published reports on root canal preparation by using the Er,Cr:YSGG laser irradiation.

Objective: The purpose of this study was to evaluate morphologically the dental hard tissue ablation at the class V cavity preparation by two types of laser devices: Er:YAG and Er,Cr:YSGG lasers in vitro.

Background Data: There have been no reports on the comparative study of dental tissue ablation at cavity preparation by Er:YAG and Er,Cr:YSGG lasers.

Methods: Twenty-two extracted human mature molar teeth were used in this study and divided into two groups of 11 teeth each.

Effects of diet and exercise on muscle and liver intracellular lipid contents and insulin sensitivity in type 2 diabetic patients.

Insulin resistance is associated with the circulating free fatty acid (FFA) level and intracellular lipid content in muscle and liver. We investigated the effect of 2-wk diet and exercise therapy on total adiposity, circulating FFA, intracellular lipid content in muscle and liver, and peripheral insulin sensitivity. Type 2 diabetic patients were divided into a diet group (n = 7) and a diet plus exercise group (n = 7).

Expression of twist and wnt in human breast cancer.

Objective: This study was performed to investigate the temperature rises on root surfaces and morphological changes on root canal walls of extracted human teeth after irradiation by erbium, chromium: yttrium, scandium, gallium, garnet (Er, Cr:YSGG) laser and to evaluate the efficiency in removing smear layer and debris from the prepared root canal walls in vitro.

Background Data: No report on Er, Cr:YSGG laser effects under various conditions and specific tips for endodontic treatments has been reported.

Methods: Sixty extracted human teeth with single and straight root were used in this study.

An economic evaluation of recombinant bovine somatotropin approval in Japan.

A comprehensive econometric model was developed to evaluate potential impacts of recombinant bovine somatotropin (rbST) approval in Japan. Three novel features of the analyses include 1) investigation of impact of rbST on herd-size structure, 2) determination of economic feasibilities of rbST adoption by herd size, and 3) evaluation of policies to mitigate negative effects of rbST approval. Simulation analysis was conducted over a 10-yr projected period assuming rbST was approved in Japan in 2001.

IV-DSA is a new diagnostic tool for axillary lymph node metastasis in breast cancer patients.

Background And Objective: The aim of this study is to know whether intravenous digital subtraction angiography (IV-DSA) is useful to detect axillary lymph node metastasis of breast cancer and to evaluate the anigiogenesis of lymph nodes in the axilla.

Patients And Methods: Forty three primary breast cancer patients (N0: 26 cases, N1: 5 cases, N2: 2 cases) who underwent IV-DSA between January and November 2000 were included in the study. Infinix CB apparatus (Toshiba, Japan) was used to collect IV-DSA images and when a mass became stained in the axilla, it was considered to be metastatic.

A close association between alteration in growth kinetics by neoadjuvant chemotherapy and survival outcome in primary breast cancer.

Few surrogate markers are available for predicting the survival benefit from chemotherapy in primary breast cancer. We examined tumor growth kinetics by assessing cytokeratin 18 neo-epitope (CK18NE), an apoptosis marker detected by M30 antibody and Ki-67 antigen, a proliferation marker detected by MIB-1 antibody in 72 primary breast cancer patients who underwent pre-operative anthracycline-based chemotherapy. Increase in M30 index and decrease in MIB-1 index after the exposure of 2 to 4 cycles of chemotherapy correlated significantly with pathological tumor response.

[Clinical usefulness of UFT and cyclophosphamide as salvage chemotherapy for patients with metastatic breast cancer].

A combination of UFT and cyclophosphamide (CPA) was orally administered to 12 women with previously-treated advanced or recurrent breast cancer. The daily dose of UFT was 400 to 600 mg for tegafur and 100 mg for CPA. Treatment was given orally for 2 weeks followed by 1 week of no treatment, or for 5 days followed by 2 days of no treatment.

Objective: The present study aimed to compare carious dentin removal by air turbine, Carisolv and erbium,chromium:yttrium,scandium,gallium,garnet (Er,Cr:YSGG) laser, and examine morphological changes before and after these caries removal techniques under light microscopy and scanning electron microscopy (SEM).

Background Data: Although there have been numerous studies on removing caries by Er,Cr:YSGG laser, none has compared Er,Cr:YSGG laser and Carisolv, or reported on the usage of DIAGNOdent as a diagnostic tool particularly for advanced caries in in vitro experiments.

Materials And Methods: Sixty extracted human teeth diagnosed as advanced caries were divided into three groups based on the treatment received, namely air turbine, Carisolv, and Er,Cr:YSGG laser groups.

A case of diabetic mastopathy with multiple lesions mimicking breast cancer.

We report a case of diabetic mastopathy with multiple unilateral lesions in an insulin dependent patient. The patient was a 62-year-old woman with two hard tumors in the right breast, who had been treated with insulin for diabetes mellitus. Mammography revealed a highly dense tumor in the right breast, while ultrasonography showed two irregular hypoechoic lesions with marked posterior acoustical shadowing, suggesting scirrhous carcinoma.

A patient with hyperlipidemia who had a gallbladder stone caused by administration of fenofibrate.

Fibrates are widely used for treatment of hyperlipidemia. It has been reported that gallbladder stones are formed by administration of clofibrate. It is thought that fenofibrate can cause the formation of gallbladder stone as a side effect.

Expression of cyclooxygenase-2 in malignant and benign breast tumors.

Cyclooxygenase-2 (COX-2) is reported to play an important role in carcinogenesis. We examined COX-2 expression in patients with breast cancer and benign breast tumors. Immunohistochemical staining revealed a high level of COX-2 expression in malignant lesions of invasive ductal carcinoma (IDC) at a rate of 40% and a low level of expression in 15% of adjacent normal-appearing breast epithelia.

With the increased knowledge obtained from more formal and rigorous clinical trails in recent years on the benefit of more intensive management of blood glucose and blood pressure in preventing or delaying the development and progression of diabetic micro- and macro-angiopathies, the challenges for routine clinical practice have become even greater. The complexity of the condition has also become evident, with realization that control of glucose, lipid and blood pressure levels, are all primary management goals.

Dual subcellular distribution of cytochrome b5 in plant, cauliflower, cells.

Subfractionation studies showed that cytochrome b(5) (cyt b5), which has been considered to be a typical ER protein, was localized in both the endoplasmic reticulum membrane (ER) and the outer membrane of mitochondria in cauliflower (Brassica olracea) cells and was a component of antimycin A-insensitive NADH-cytochrome c reductase system in both membranes. When cDNA for cauliflower cyt b5 was introduced into mammalian (COS-7) and yeast cells as well as into onion cells, the expressed cytochrome was localized both in the ER and mitochondria in those cells. On the other hand, rat and yeast cyt b5s were specifically localized in the ER membranes even in the onion cells.

Integration of cytochrome b5 into endoplasmic reticulum membrane: participation of carboxy-terminal portion of the transmembrane domain.

Integration of cytochrome b(5) (b5), a tail-anchored protein located in the endoplasmic reticulum (ER) membrane, into the membrane was studied. Mutation of three amino acids, -Leu-Met-Tyr, at the carboxy-terminal end of the transmembrane segment of b5 to alanines resulted in localization of the mutated protein, b5LMY/AAA, in the cytosol as well as in the ER membrane. When an N-glycosylation site was introduced at the carboxy-terminal end of b5LMY/AAA, a substantial amount of the glycosylated form of the mutant protein was recovered in the cytosol fraction.

Histological classification of invasive ductal carcinoma and the biological parameters in breast cancer.

Background: The histological classification of invasive ductal carcinoma proposed by the Japanese Breast Cancer Society is based on the appearance of breast cancer invasion. However, few studies have been done to clarify the relationship between the histological classification and other parameters that represent the biological characteristics of individual breast cancer cells.

Materials And Methods: We analyzed 1,025 invasive ductal carcinomas, consisting of 424 papillotubular, 330 solid-tubular and 271 scirrhous cases, with regard to the status of estrogen receptor (ER), progesterone receptor (PgR), DNA ploidy, epidermal growth factor receptor (EGFR) and p53 protein.

Adenocarcinoid of the appendix: report of two cases.

Adenocarcinoid of the appendix is a rare tumor with the histological features of both adenocarcinoma and carcinoid tumor. However, its biological behavior and malignant potential are still unclear. We treated two patients with this unusual tumor; a 60-year-old man and a 79-year-old woman.

Identification of outer mitochondrial membrane cytochrome b5 as a modulator for androgen synthesis in Leydig cells.

Outer mitochondrial membrane cytochrome b5 is an isoform of microsomal membrane cytochrome b5. In rat testes the outer mitochondrial membrane cytochrome b5 is present in both mitochondria and microsomes, whereas microsomal membrane cytochrome b5 is undetectable. Outer mitochondrial membrane cytochrome b5 present in the testis was localized in Leydig cells with cytochrome P-45017alpha, which catalyzes androgenesis therein.
