Antisense oligonucleotides (ASOs) are an important class of therapeutics to treat genetic diseases, and expansion of this modality to neurodegenerative disorders has been an active area of research. To realize chronic administration of ASO therapeutics to treat neurodegenerative diseases, new chemical modifications that improve activity and safety profiles are still needed. Furthermore, it is highly desirable to develop a single stereopure ASO with a defined activity and safety profile to avoid any efficacy and safety concerns due to the batch-to-batch variation in the composition of diastereomers.

Background Prior to using the exoscope, we speculated that it represented an intermediate tool between a loupe and a microscope and had concerns about its visibility of deep, fine structures. Objective To evaluate the depths of meningioma for which the exoscope was suitable, and to clarify its disadvantages in meningioma resection. Methods Findings of consecutive meningioma surgeries using a 4K three-dimensional (3D) exoscope over a one-year period were evaluated for visibility of the surgical field, comfort of the surgeon's arm posture, the surgeon's head orientation, and perception of the image delay, accounting for the depth of the tumor.

Soft tissue and bone tumors are rare, and their low frequency and diverse histological types make conducting large-scale clinical trials challenging. Patient-derived xenografts (PDX), entailing implantation of cancer specimens in immunocompromised mice, are emerging as a valuable translational model because PDX keeps the original tumors' character and drug sensitivity. We sequentially transplanted 166 surgical and biopsy specimens from orthopedic surgeries, including 138 soft tissue and bone tumors (81 malignant, 23 intermediate, and 34 benign), 16 metastatic bone tumors, 9 hematological malignancies, and 3 non-tumor tissues.

Article Synopsis
  • Biological neural networks, especially in the visual cortex of different species, process information through both spontaneous and stimulus-evoked activities, but how they handle internal noise vs. sensory signals is debated.
  • In mouse primary visual cortex (V1), spontaneous and stimulus-evoked activities reveal dissimilar patterns, aiding in the separation of sensory information from noise, while higher similarity is observed in carnivores and primates.
  • The study found that in marmosets, while V1 shows similar spontaneous and stimulus-evoked activity, higher visual areas exhibit an increasing orthogonalization of these patterns, suggesting a broader principle of cortical computation across species.

  • Endoscopic pancreatic stenting (EPS) is effective for treating pain in chronic pancreatitis, but there's limited understanding of why pain recurs after stent removal and no clear guidelines for when to remove stents.
  • A study of 95 patients who underwent EPS identified serum lipase level and pancreatic parenchymal thickness as key risk factors for pain recurrence after stent removal.
  • A prediction model based on these risk factors was developed, demonstrating good ability to predict pain recurrence which could aid in making informed decisions regarding pancreatic stent management.

The working curve informs resin properties and print parameters for stereolithography, digital light processing, and other photopolymer additive manufacturing (PAM) technologies. First demonstrated in 1992, the working curve measurement of cure depth vs radiant exposure of light is now a foundational measurement in the field of PAM. Despite its widespread use in industry and academia, there is no formal method or procedure for performing the working curve measurement, raising questions about the utility of reported working curve parameters.

  • Sarcopenia, characterized by loss of skeletal muscle and reduced grip strength, is common in chronic pancreatitis (CP) patients, but the study explores their different impacts on health.
  • In the study of 102 CP patients, 55.9% had muscle loss, while 20.6% had reduced grip strength, revealing that muscle loss was linked to worse nutritional status and pancreatic function.
  • The findings suggest that muscle loss correlates with serious pancreatic issues like duct dilation and tissue atrophy, whereas reduced grip strength is more associated with older age and nutritional decline.

Article Synopsis
  • Over 70% of patients with neurofibromatosis type 1 (NF1) exhibit various central nervous system abnormalities, including tumors and non-neoplastic conditions.
  • In a study of three cases, lesions initially thought to be non-neoplastic turned out to be jelly-like masses during surgery, suggesting a possible neoplastic nature, despite microscopic analyses showing traits aligning with normal arachnoid cells.
  • Molecular tests revealed no definitive signs of cancer, leading researchers to propose that these lesions might represent a rare form of hyperplasia of arachnoid trabecular cells associated with NF1.
