Acute myeloid leukemia (AML) is an aggressive malignancy that requires rapid treatment with chemotherapies to reduce tumor burden. However, these chemotherapies can compromise lymphocyte function, thereby hindering normal anti-tumor immune responses and likely limiting the efficacy of subsequent immunotherapy. To better understand these negative impacts, we assessed the immunological effects of standard-of-care AML therapies on lymphocyte phenotype and function over time.

The second year of the COVID-19 (Coronavirus Disease 2019) pandemic has seen the need to identify and assess the long-term consequences of a SARS-CoV-2 (Severe acute respiratory syndrome coronavirus-2) infection on an individual's, including adequate cognitive functioning. This review is attempting to highlight the current understanding of the various direct and indirect mechanisms of nervous system damage, describing the causality of similar symptoms following SARS-CoV-2 infection. This understanding is essential to establish the long-term consequences from the disease (including the potential for increased risk of dementia in some cases) and to identify means of preventing or ameliorating the brain damage.

Abdominal aortic aneurysm (AAA) is a prevalent life-threatening disease, where aortic wall degradation is mediated by accumulated immune cells. Although cytokines regulate inflammation within the aorta, their contribution to AAA via distant alterations, particularly in the control of hematopoietic stem cell (HSC) differentiation, remains poorly defined. Here we report a pathogenic role for the interleukin-27 receptor (IL-27R) in AAA, as genetic ablation of IL-27R protects mice from the disease development.

Article Synopsis
  • - The study examined how TLR2 influences the production of reactive oxygen species (ROS) in mouse peritoneal macrophages using a specific chemiluminescence technique when stimulated by different ligands like zymosan and peptidoglycan.
  • - It was found that the levels of ROS produced by macrophages were affected by the concentration ratios of these ligands, indicating that their effects were not independent of each other.
  • - Three main interactions were identified: the combined stimulating effects of the ligands, competition between the ligands for binding, and how prior exposure to peptidoglycan primed macrophages for better response when zymosan was used.

Primary pancreatic injury that occurs in acute pancreatitis leads to necrosis of pancreatic cells and is accompanied by the development systemic inflammatory response of varying severity. Systemic inflammatory response, in turn, can lead to the development of multiple organ dysfunction syndrome and death of patients. The release of damage-associated molecular patterns into the extracellular space is the trigger pathological mechanism underlying these processes.

In this work the effect of the natural complex of cytokines on the generation of active forms of oxygen (AFO) by mouse peritoneal macrophage in different in vivo and in vitro systems were studied. Intact mice of various strains were found to have differences in the AFO production of their macrophages: BALB/c (H-2d) > CBA (H-2k) > C57BL/6 (H-2b), as well as in the degree of the prestimulation of the macrophage oxygen metabolism with the complex of cytokines BALB/c > CBA. The in vivo and in vitro infection of macrophages of mice, oppositely reacting to Leishmania donovani, led to the gradual suppression of the capacity of macrophages for AFO generation.
